Procedural improvements suggested in the use of [35S]SO4 uptake as a possible measure of microbial production include: prompt filtration and sterilization of stock solutions, use of polycarbonate Nuclepore filters, prewetting filters, addition of Na2SO4 to samples before filtration, rinsing filters with 1 g liter−1 (0.01 M) Na2SO4, removal of filter edges before liquid scintillation counting, and dissolving Nuclepore filters in phenethylamine before adding scintillation cocktail. This procedure results in control filter counts of 0–200 cpm above background at isotope concentrations of up to 2.0 µCi ml−1 of sample.
