Darier's Disease Associated with Cutaneous Malignancies

Abstract: There have been no reports of cutaneous malignancies arising in lesions of patients with Darier's disease. Two such cases are herewith reported, one a carcinoma of adnexal origin, and the other of several basal‐cell carcinomata. Possible predisposing factors are briefly discussed and speculated upon.

“…17 Recent data suggest that the ATP2A2 gene is also correlated with several cancer types in humans. [19][20][21][22] Although squamous cell carcinoma is not common in patients with DD, 7 cases of oral, subungual, vulval, and scrotal squamous carcinoma have been reported in addition to basal cell carcinoma, melanoma, and non-small cell lung cancer [23][24][25][26][27][28][29] ; in the latter case, sequencing of the ATP2A2 gene revealed 3 polymorphisms and one ATP2A2 mutation (R751Q). 20 No DD patient with cancer has been detected in our cohort; however, the majority of patients were younger than 40 years.…”

“…Since 1981, 13 cases of NMSC in patients with DD have been reported: 7 squamous cell carcinomas (SCC), 5 BCC, and 1 adenexal tumor. Of the 5 cases of BCC, 2 of the cases had previously received grenz-ray therapy and/or superficial radiotherapy [12, 13], while no risk factors were associated with the other 3 cases [13–15] (Table 2). …”

“…Darier's disease has not been associated with cutaneous malignancies. In 1981, Latour et al 1 reported two patients with Darier's disease who developed skin cancers: one developed an adnexal carcinoma and the other developed multiple BCCs. The second patient had received superficial radiotherapy and grenz‐ray therapy.…”

“…Darier's disease has not been found to be associated with cutaneous malignancies. In an extensive literature review only three patients were reported with Darier's disease and cutaneous malignancies; two cases had basal cell carcinoma (BCC) and one had an adnexal carcinoma 1 (Table 1). A patient with Darier's disease who developed multiple BCCs is presented.…”
