Daily Variations of Homocysteine Concentration May Influence Methylation of DNA in Normal Healthy Individuals

Abstract: The regulation of genetic expression is tightly controlled and well balanced in the organism by different epigenetic mechanisms such as DNA methylation and histone modifications. DNA methylation occurring after embryogenesis is seen mainly as an irreversible event. Even small changes in genomic DNA methylation might be of biological relevance, and several factors influencing DNA methylation have been identified so far, one being homocysteine. In this study, genomic DNA methylation was analyzed and homocysteine… Show more

“…In our data, LINE-1 and Alu methylation had associations with mean outdoor temperature of the day of the exam that, although not significant, were consistent with those observed with season. A previous study of 30 healthy volunteers reported cyclic variations in blood DNA methylation content observed over a 24-hour period (36), and recent investigations have shown transient cyclical methylation and demethylation of specific gene promoters (37,38). The association of LINE-1 methylation with day of the week observed in our data may provide further evidence for the existence of cyclical patterns of variation in repetitive element DNA methylation.…”

“…A previous study that assessed global DNA methylation by fluorescently labeled cytosine extension assay reported diurnal changes in global DNA methylation in humal blood. 50 The present study is the first to demonstrate a diurnal rhythm of global DNA methylation in the liver. Furthermore, the variation appeared to be coordinated with the diurnal variation in Dnmt3b expression.…”

“…In humans, several peptide hormone levels show daily variations; growth hormone increases during sleep (19), leptin increases during the evening (20), and prolactin increases at night (21). Concentrations of amino acids, including tryptophan, tyrosine, phenylalanine (22), methionine (23), cysteine, glutathione (24), and homocysteine (25), also exhibit daily variations in human blood plasma. Despite these findings, comprehensive profiling of circadian dynamics of chemical substances in mammalian blood has not yet been reported, and until now a comprehensive molecular timetable of such chemical substances has not been constructed.…”
