d-Serine and a glycine transporter-1 inhibitor enhance social memory in rats

Abstract: The present results show that stimulation of the glycine modulatory sites on the NMDA receptor either directly with D: -serine or by blocking glycine transporter-1 enhances social memory and may be an effective approach for the treatment of the cognitive dysfunction observed in schizophrenic patients.

“…1A, 2A, and 3), indicating that MK-801 impaired short-term social memory, as we previously reported (16). The administration of the mGlu2/3-receptor agonist LY379268 and the mGlu2-receptor potentiator BINA significantly improved the social memory impairment elicited by MK-801 in rats {[F(2,32) = 5.51, P < 0.01] and [F(2,43) = 4.75, P < 0.05] (Figs.…”

“…Exogenous administration of D-serine leads to improvements in specific features of spatial memory in the Morris water maze, such as reversal learning (Duffy et al 2008), probe test retrieval (Zhang et al 2008), and day-to-day savings in the delayed-match-to-place version of the task (Stouffer et al 2004). In addition, D-serine administration improves social memory in rats (Shimazaki et al 2010). However, most of the above cited studies used doses of 600-1,300 mg/kg (with the exception of that by Stouffer et al 2004, which used 100 mg/kg), far above the range shown to be effective in clinical trials with schizophrenia patients (30-120 mg/kg).…”
