Oxidative stress in the body occurs when the production of free radicals overwhelms the antioxidant defence systems. The aim of this study was to examine the effects of a tiletamine/ zolazepam-xylazine-tramadol combination on the antioxidant status, lipoperoxidase and blood cell indicators in eight miniature pigs. Baseline measures were recorded prior to anaesthesia, then the animals were anaesthetized with the combination of tiletamine/zolazepam (3.5 mg/kg), xylazine (1.32 mg/kg), and tramadol (1.8 mg/kg). Blood samples were collected from the anterior vena cava at 15, 30, 60, 90, 120 min, 24 h, and 72 h after anaesthesia. Plasma malondialdehyde, superoxide dismutase, catalase, and glutathione peroxidase concentrations were measured by colorimetry, and red blood cell counts, white blood cell counts, haemoglobin and packed cell volume were determined using an automated cell counter. The results showed that the concentration of malondialdehyde increased significantly at 30 and 60 min after the injection (P < 0.05), whereas glutathione peroxidase and catalase activity increased slightly (P > 0.05) then returned to baseline values after 90 min (P > 0.05). Superoxide dismutase activity increased significantly (P < 0.05) at 30 and 60 min, and then gradually decreased to baseline values after 90 min. Changes in red blood cell counts, haemoglobin and packed cell volume were not significant, while white blood cell count decreased significantly (P < 0.05) at 30 and 60 min. Our study is the first to demonstrate that tiletamine/zolazepam-xylazine-tramadol provide antioxidant effects, which may be proposed for alleviating the stress of examination and research at veterinary clinics or long-distance transportation.

A AB BS ST TR RA AC CT T O Ob bj je ec ct ti iv ve e: : This study aimed to investigate the total oxidant status (TOS), total antioxidant status (TAS), oxidative stress index (OSI) and the paraoxonase and arylesterase activities of patients with gallstones and compare them with healthy individuals as well as to assess the impact of sevoflurane and desflurane on oxidative stress markers in laparoscopic surgery. M Ma at te er ri ia al l a an nd d M Me et th ho od ds s: : Forty patients scheduled for laparoscopic surgery (Patient Group) and 30 healthy volunteers as control subjects (Control Group) were enrolled in the study. The patient group was randomized to desflurane group (n= 0) and sevoflurane group (n= 20). Blood samples were collected preoperatively and at postoperative 6 hours in order to measure the levels of TAS, TOS, OSI, and paraoxonase and arylesterase activities. Preoperative and postoperative laboratory findings of sevoflurane and desflurane group were compared with the Mann Whitney U test between groups and the Wilcoxon test within a group. R Re es su ul lt ts s: : Preoperative TOS, OSI levels in the patient group were significantly increased and arylesterase activities were significantly decreased compared to the control group (p<0.05) [(TOS (µmol H2O2 equiv/L), 19.64 ± 2.16 vs. 14.25 ± 1.83), (OSI 1.85 ± 0.53 vs. 0.98 ± 0.24), (Arylesterase (U/L), 45.19 ± 6.82 vs. 49.66 ± 4.78)]. There was no statistically significant difference between sevoflurane and desflurane group in laboratory findings of the preoperative and the postoperative period (p> 0.05). Preoperative and postoperative TAS, TOS, OSI results and activities of paraoxonase and arylesterase were not significantly different in the sevoflurane group (p> 0.05). Although the preoperative and postoperative levels of TAS and TOS and paraoxonase and arylesterase activities in the desflurane group was not significantly different, postoperative OSI was significantly increased (p< 0.05) (OSI 1.80 ± 0.57 vs 2.20 ± 0.67). Summary: This study was designed to investigate the effects of low-flow anesthesia with sevoflurane and desflurane on renal and hepatic functions in patients undergoing laparoscopic abdominal surgery. Twenty patients with ASA I or II (American Society of Anesthesiologists classification) physi cal scores were included in the study. There were no signifi cant differences between sevoflurane and desflurane groups with respect to age, weight, body mass index, duration of the operation and the anesthesia. In both groups, renal function parameters such as urea, BUN, creatinine and calculated creatinine clearance did not show significant differences at 24 and 48 hours. Homocysteine levels, which showed renal metabolic function, did not change significantly at 24 and 48 hours when compared to baseline le vels in both groups. Transaminases were not significantly different between the two groups from baseline to 24 and 48 hours. These differences between the preoperative and post operative values of biochemical parameters were similar for both anes thetic groups (p>0.05). Low-flow anesthesia did not cause impairment in terms of renal and hepatic functions.
