Cytotoxic Effects of Newly Synthesized Palladium(II) Complexes of Diethyldithiocarbamate on Gastrointestinal Cancer Cell Lines

Abstract: As a part of a drug development program to discover novel therapeutic and more effective palladium (Pd) based anticancer drugs, a series of water-soluble Pd complexes have been synthesized by interaction between [Pd (phen)(H2O)2(NO3)2] and alkylenebisdithiocarbamate(al-bis-dtc) disodium salts. This study was undertaken to examine the possible cytotoxic effect of three novel complexes (0.125–64 µg/mL) on human gastric carcinoma (AGS), esophageal squamous cell carcinoma (Kyse-30), and hepatocellular carcinoma (H… Show more

“…The obtained IC 50 values are 1.5, 1.8, 3.4 and 6.1 mM for 1 , 2 , Met and IDA, respectively. These values of the ligands and their Ni (II) complexes were compared to cisplatin and were found to be higher as compared to those achieved for cisplatin (IC 50 = 0.2 mM) . The trend of cytotoxicity was found to be complex 1 < complex 2 < Met < IDA.…”

“…The metal chelate interacts with DNA, likely in a mode that includes electrostatic or hydrophobic interaction. As expressed previously, the CuL complex has two replaceable water molecules coordinated to copper(II) ion in solution, which might be substituted with DNA double helix molecules in the solution [35,41,45,58,59]. Likewise, because of the replacement reaction of water ligand molecules from the CuL complex in solution, the explored metal chelates would have a flat part in the middle.…”

“…Among all, tetrazolium (MTT) is one of the most often used, which uses colorimeter to determine cell viability. Cytotoxicity test is done to assess cell viability by a dose response compounds' toxicities [104106]. Cytotoxicity is usually assessed with MTT [3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide] assay.…”
