A class of dietary fibers and biologically active polysaccharides from natural sources, beta-glucans (βTGs) have bioactive capabilities. The anti-tumor, anti-inflammatory, prebiotic, anti-obesity, anti-allergic, anti-microbial, antiviral, anti-osteoporotic, and immunomodulating effects of βTGs are well documented. Although many biological activities of βTG have been proven, its mechanism in DHEA-induced PCOS has not been investigated. We aimed to investigate the protective effects of βTG treatment on PCOS and its capacity to reverse PCOS-induced changes. Female Sprague-Dawley (SD) rats were divided into four groups at random (n = 8): control, PCOS, PCOS + βTG, and βTG groups. Biochemical markers linked to oxidative stress, antioxidant state, inflammation, cytokines, and hormone levels were assessed. Analyses using immunohistochemistry and histopathology were also carried out. Membrane array analysis detected growth factors, cytokine, and chemokine protein profiles. βTG did not cause any change in body, uterus, and ovarian weights in rats. βTG normalized the deviations in the oestrus cycle caused by PCOS. It was observed that βTG had a positive effect on the reproductive system. βTG can reduce the inflammatory response in PCOS rats by decreasing inflammatory cytokines. Oxidative stress was significantly reduced, whereas antioxidant enzyme activities were significantly elevated in the βTG group. βTG also prevented histopathological alterations. βTG induced the expression of some essential proteins, including bNGF, TIMP-1, Agrin, CINC-1, BDNF, and FGF-2 (bFGF). The results of this study showed that treatment with βTG protects against oxidative stress, inflammation, hormone imbalance, and histopathological damage in ovarian tissue caused by PCOS.

Bu çalışma kapsamında, halk arasında acı kavun olarak bilinen Ecballium elaterium'un, bazı kanser hücre hatlarına karşı sitotoksik etkilerinin araştırılması hedeflenmiştir. Eski Mısırlılar döneminden beri tanınan bir drog olan E. elaterium birçok hastalığa karşı kullanılmıştır. Bu çalışmada ise son derece saldırgan ve tedavisi bulunmayan malign melanoma (SK-MEL-30), kolon kanseri (CACO-2), meme kanseri (MCF-7) ve küçük hücreli olmayan akciğer kanseri (H1299) hücre hatlarına karşı E. elaterium'un meyve ve yaprak ekstrelerinin sitotoksik etkileri MTT testi ile değerlendirilmiştir. E. elaterium'un meyve ve yaprak ekstreleri aynı zamanda sağlıklı fare fibroblast hücre hattı (L929) üzerinde denenmiştir. Sonuç olarak; E. elaterium'un meyve ve yaprak ekstreleri; SK-MEL-30 hücre hattına karşı zayıf, H1299 hücre hattına karşı orta, MCF-7 hücre hattına karşı yüksek sitotoksik aktivite sergilemiştir. CACO-2 hücre hattına karşı ise her iki ekstrenin de toksik bir etkisi olmadığı gözlemlenmiştir. Ayrıca, meyve ekstrelerinin yaprak ekstrelerinden daha yüksek sitotoksik aktivite sergilediği tespit edilmiştir.

Background: Cyclophosphamide (CP) is one of the chemotherapeutic and immunosuppressive agents. It has multiple toxic effects, particularly hepatotoxicity. Ecballium elaterium (EE) plant is found in the Mediterranean counties. It has been used as a traditional medicine. Objectives: This work aimed to evaluate the ameliorative effect of EE fruit juice on hepatotoxicity induced by CP in rats. Material and methods: 28 male rats were divided into equal 4 groups: control group, EE group (0.2 mL/kg orally for 10 days), CP group (2 doses of 150 mg/kg intraperitoneally in the first 2 days), and EE plus CP group. Biochemical analysis for serum levels of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), total bilirubin and albumin were carried out. Hepatic oxidant/antioxidant markers; glutathione (GSH), glutathione reductase (GR), superoxide dismutase (SOD), catalase (CAT), and lipid peroxidation product; malondialdehyde (MDA) in addition to pro-inflammatory mediators; tumor necrosis factor-α (TNF-α), interleukin (IL-1β) and IL-6 were evaluated. Histopathological examination of the liver was investigated. Results: Serum liver enzymes and total bilirubin were significantly increased with a decrease in albumin in the CP treated group compared to control group. CP induced depletion of oxidative stress defensive pathways with increased MDA levels. The examined pro-inflammatory mediators were significantly elevated as compared to control group with disruption of hepatic histopathology. All these hepatic disturbances were ameliorated by administration of EE fruit juice with CP. Conclusion: Administration of EE fruit juice ameliorates the hepatotoxicity induced by CP through its antioxidant and antiinflammatory activity.
