Cytotoxic Arylnaphthalene Lignans from a Vietnamese Acanthaceae, Justicia patentiflora

Abstract: One new norlignan (1) and five new lignans (2-6) were isolated from the leaves and stems of Justicia patentiflora by a bioassay-guided purification. Five known compounds, carinatone, diphyllin, justicidin A, taiwanin E, and tuberculatin, were also found in J. patentiflora. Most of the new compounds display significant activity in in vitro cytotoxic assays against KB, HCT116, and MCF-7 cancer cell lines and arrest the cell cycle in the G0/G1 phase.

“…The MS/MS spectra of peak 21 gave product ions by the loss of H 2 O, CH 3 radical, propene, CO, C 4 H 8 . Thus, 15, 20 and 21 could only be tentatively assigned as 3-oxoisotaxodione [53], 6-hydroxysalvonolone [54] and miltirone [30,31], respectively. 15 and 20 are actually two marker components that differentiate S. dugesii Fernald (sample 10) and S. roborowskii (sample 23) Maxim from other Salvia species.…”

“…Daurinol is isolated from a traditional ethno-pharmacologic plant, Haplophyllum dauricum, which has historically been used to treat tumors in Russia (33). Antiproliferative activities of arylnaphthalene lignans against various human cancer cells have been reported, but the molecular mechanism of their antiproliferative effects is still poorly understood (34)(35)(36). Daurinol displayed strong antitumor activity without any significant adverse effects (36).…”

“…4''-O-Acetylmananthoside B was first discovered and reported by Guéritte and coworkers, 1 which was isolated from the leaves and stems of Justicia patentaflora in Vietnam and displays significant cytotoxicity ( Figure 1). However, mananthoside B, possessing an almost identical molecular skeleton, was first disclosed by Zhang and coworkers, 2 which also shows cytotoxicity ( Figure 1).…”
