Chemotaxonomy in the Etlingera Genus from Sulawesi, Indonesia: Design and molecular docking of antioxidant marker. Biodiversitas 25: 449-457. Etlingera is one of the important genera in the Zingiberaceae family because of its potential uses. Several studies have been conducted on the Genus Etlingera as nutraceuticals and drugs. Recently new species have been found in the Sulawesi Region, Indonesia, namely E. canaria A.D.Poulsen and E. echinulate A.D.Poulsen. Based on literature review through sciencedirect and springer link, both species unknown for their chemical and pharmacological content, so it is necessary to continue to reveal the deeper potential. The fruits of E. calophrys (K.Schum.) A.D.Poulsen, E. canaria and E. echinulate were extracted with methanol by maceration method. The chemical content of each extract was analyzed using a Liquid Chromatography Mass Spectrometer (LCMS). Antioxidant activity was tested based on the 1,1-Diphenyl-2-Picrylhydrazyl (DPPH) method. The ability as an antioxidant is proven by the molecular interactions of marker compounds in inhibiting xanthine oxidase. The secondary metabolites contained in the fruit extracts of E. calophrys, E. canaria and E. echinulate are E-p-Coumatic acid, 1,5-Dimethyl citrate, Trimethyl citrate, Crocetin, Tanshinone IIA, Ethinyl Estradiol, Yakuchinone B, Anisalacetone, Yakuchinone A, Nobilin, ?-Estradiol, Momor-cerebroside I, Spinasterone, 12-Hydroxy-methyl abietate, Spinasterol, 2,3-Dihydroxypropyl oleate, 11-Octadecenoate acid methyl ester, Stigmastan-3,6-dione, b-Sitosterol- 3-O-b-D-glucopyranoside, Trilaurin, 28-O-Hexahydrophthalate and Fasciculol C. Yakuchinone A and Yakuchinone B are chemotaxonomic marker compounds identified in all species. The antioxidant properties of each extract of 42.27±0.53, 46.59±0.81 and 35.66±0.73 mg/L have been proven by the molecular interactions of the marker compounds in their ability to act as xanthine oxidase inhibitors. The compounds Yakuchinone A and Yakuchinone B are thought to be responsible for the pharmacological activity of the species E. calophrys, E. canaria and E. echinulate. Yakuchinone A and Yakuchinone B are marker compounds because they are distributed in all samples.

Objective. Lung cancer represents the leading cause of cancer death on a global scale. Gynostemma pentaphyllum (G. pentaphyllum), a traditional medicinal material with a high medicinal and health value, has recently been reported for its anticancer activity. However, the pharmacological mechanism of G. pentaphyllum in non-small-cell lung cancer (NSCLC) remains to be elucidated. Methods. The active ingredients of G. pentaphyllum were obtained from the TCMSP database and known therapeutic targets of NSCLC from the GeneCards and OMIM databases. Disease-drug common targets are subjected to protein-protein interaction (PPI), GO enrichment analysis, and KEGG pathway enrichment analysis. A molecular docking strategy was performed to verify the interaction between molecules. Results. We found a total of 24 compounds of G. pentaphyllum fulfilling OB ≥ 30% concomitant with DL ≥ 0.18 and corresponding 81 target genes in the TCMSP database, with 5062 NSCLC-related genes collected in the GeneCards and OMIM databases. The network consisting of the disease-target compound was obtained, including 8 active ingredients and 69 common targets. The PPI network with 65 nodes and 645 edges was visualized. After functional enrichment analysis, it was revealed that the therapeutic effects of G. pentaphyllum on NSCLC were achieved through response to ketone, gland development, and cellular response to xenobiotic stimulus. After molecular docking analysis, it was revealed that the two active ingredients of G. pentaphyllum, quercetin and rhamnazin, bound well and stably to their targets (MYC, ESR1, and HIF1A). Conclusion. Our study, based on network pharmacology, identifies active ingredients, targets, and pathways model mechanism of G. pentaphyllum when it is used to treat NSCLC.

Background: Etlingera rubroloba (E. rubroloba) A.D Poulsen is one of the endemic plants of Southeast Sulawesi, which has the largest species in the world. Local people widely use this plant as traditional medicine. Many of these plant species have been previously reported as antioxidants, antibacterials, and others. However, for E. rubroloba, there are still very few reports from a pharmacological aspect, especially anthiperuricemia. Objective: This study was conducted to determine the characteristics and activity of the ethanol extract of the stem of E. rubroloba A.D Poulsen in vitro. Methods: The stems of E. rubroloba A.D Poulsen were extracted with ethanol solvent by maceration, then the filtrate obtained was evaporated using an evaporator to get an ethanol extract. Furthermore, the extract characterization test was carried out, including water content, ash content, water-soluble extract content, and ethanol-soluble extract content. Then the antihyperuricemia activity test in vitro using the Xanthine Oxidase (XO) kit. Results: The results of the characteristic test showed that the ethanolic extract of E. rubroloba contained 5.15% water content, 5.485% ash content, 55.2% water-soluble extract content, and 56.52% ethanol-soluble extract content. Furthermore, the antihyperuricemic activity of the ethanol extract of E. rubroloba was IC50 48.59 ± 0.03 g/mL and Allopurinol 42.21 ± 0.02 g/mL as positive controls. Conclusion: E. rubroloba stem ethanol extract has characterization results according to the literature, has antihyperuricemia activity, and is a reference for the development of traditional drugs
