Cytotoxic activity and T cell receptor repertoire in tumor-infiltrating lymphocytes of adrenal cell carcinomas

Abstract: The cytotoxic activity and T cell receptor (TCR) V beta repertoire in tumor-infiltrating lymphocytes (TIL) of three primary adrenal cell carcinomas were analyzed. Fresh, non-cultured TIL from two of the three tumors showed low but significant lysis of the autologous tumor, and for one of the patients this activity was strongly enhanced upon culture in interleukin-2. An allogeneic adrenal cell carcinoma line and the K562 or Daudi targets included as controls were not killed. Phenotypic analysis of freshly isola… Show more

“…This included tumors such as Wilms tumor, adrenal gland carcinoma where none of those we examined surpassed the 10 CPM cutoff. This level conservatively suggested that less than 1 in 100,000 cells were infected with EBV presuming viral miRNAs were 25% of total cellular content 19,20 .…”

“…The approaches to the study of the TCR repertoire have largely been conducted on monoclonal populations of T cells using either reverse transcriptase-PCR-based techniques or flow cytometry (20,27,28). Examination of monoclonal populations of T cells usually provides unambiguous information concerning expression of V␤ at the single cell level.…”

“…To further investigate the role of oxidative stress in relation to tumor immunology, we have examined how oxidative stress affected different subsets of T cells, with a particular focus on the cytokine production by cells expressing the memory/activation marker CD45RO, as this marker is commonly expressed by T cells infiltrating into tumors (11)(12)(13). We have also attempted to delineate whether loss of T cell function after H 2 O 2 exposure is a predecessor to apoptosis of T cells or an early consequence following the initiation of the apoptotic process.…”
