Cytosolic Glyceraldehyde-3-Phosphate Dehydrogenase Is Phosphorylated during Seed Development

Abstract: Cytosolic glyceraldehyde-3-phosphate dehydrogenase (NAD-GAPDH) is involved in a critical energetic step of glycolysis and also has many important functions besides its enzymatic activity. The recombinant wheat NAD-GAPDH was phosphorylated in vitro at Ser205 by a SNF1-Related protein kinase 1 (SnRK1) from wheat heterotrophic (but not from photosynthetic) tissues. The S205D mutant enzyme (mimicking the phosphorylated form) exhibited a significant decrease in activity but similar affinity toward substrates. Immun… Show more

“…For example, S -glutathionylation of TRX f in chloroplast could alter its oxido-reductase activity toward targets, affecting them indirectly (Michelet et al, 2005). The SnRK1 is an essential regulator of plant metabolism by direct phosphorylation of a number of enzymes involved in carbohydrate metabolism and nitrogen assimilation (Piattoni et al, 2017; Wurzinger et al, 2018). Recently, it was determined that A. thaliana SnRK1 kinase activity is sensitive to inhibition by H 2 O 2 .…”

“…Phosphorylated α-GP is active, and dephosphorylated α-GP is inactive [43]. GAPDH catalyzes a critical step in glycolysis; GAPDH activity was significantly decreased if phosphorylated [44]. In this study, phosphorylated α-GP was up-regulated both in CO39 and C101LAC 12 h after SA treatment, suggesting that SA might increase α-GP activity.…”

“…Starch content was determined with the anthrone method [ 93 ] at 640 nm using soluble starch as standard. For protein analysis, 0.5 g of dry seeds were ground and extracted according to the method as described by Piattoni et al [ 94 ]. The total protein content was measured by using a spectrophotometer at 595 nm following the Bradford protocol [ 95 ].…”
