Cytosolic accumulation of γH2AX is associated with tropomyosin-related kinase A-induced cell death in U2OS cells

Abstract: Tropomyosin-related kinase A (TrkA) plays an important role in cell survival, differentiation, and apoptosis in various neuronal and nonneuronal cell types. Here we show that TrkA overexpression by the Tet-On system mimics NGF-mediated activation pathways in the absence of nerve growth factor (NGF) stimulation in U2OS cells. In addition, p53 upregulation upon DNA damage was inhibited by TrkA, and p21 was upregulated by TrkA in a p53-independent manner. TrkA overexpression caused cell death by interrupting cell… Show more

“…After L1210 tranfectants were treated with 1 μg/ml DXR for 24 h, the cells were treated with RNase A (250 μg/ml) and propidium iodide (50 μg/ml) for 30 min as described previously (Jung et al, 2008). Analysis of cellular DNA contents of propidium iodide-stained nuclei was measured by fluorescence-activated cell sorter (FACS; Becton Dickinson).…”

“…The neuronal SK-N-MC or non-neuronal U2OS cells expressing TrkA exhibited morphological alterations to neuron-like cells and furthermore committed to die at 12 h after addition of tetracycline. 2,3 This TrkA-induced cell death is associated not only with apoptosis but also autophagy. Overexpression of TrkA in U2OS cells causes ERK activation leading to cell cycle arrest via p21, caspase-7 activation and production of truncated BAX, directing cells to be apoptotic.…”
