Cytoskeletal organization following cannabinoid treatment in undifferentiated and differentiated PC12 cells

Abstract: Confocal microscopy in association with three-dimensional reconstruction was used to examine the changes in the microtubules and microfilaments following cannabinoid treatment of PC12 cells. Microtubules and microfilaments were disrupted in a dose-dependent manner following treatment with 10-30 microM delta 9-tetrahydrocannabinol (THC). A disruption of microtubules and microfilaments was observed following treatment with 30 microM cannabidiol and cannabinol. The amount of microtubules and microfilaments was re… Show more

“…This may be linked to structural changes or remodeling occurring after D 9 -THC exposure in adolescents and is consistent with observations of cytoarchitectural changes occurring with cannabinoid treatment (Tagliaferro et al, 2006) and other reports of altered expression of the structural-related proteins tubulin and actin (Tahir et al, 1992;Wilson et al, 1996). MBP is a major constituent of the myelin sheath that allows fast saltatory conduction of nerve pulses (Vanrobaeys et al, 2005).…”

“…These cells have been well characterized and have become a model for many studies related to identification and characterization of neuronal signaling pathways (Heneka et al, 1998), to study cell death (Dubreuil et al, 2003;Kitazawa et al, 2004;Ulloth et al, 2003;Yang et al, 2004), especially apoptosis, in neuronal cells and cytoskeletal structure and function (Kobayashi and Mundel, 1998;Nakayama et al, 2001;Okabe and Hirokawa, 1988;Tahir et al, 1992). Although no other types of cells were examined in the present study, cell viability studies and onset rate dependency of traumatic injury produced on cells with CCSD were similar to previous studies conducted with human derived neuronal cells (Ellis et al, 1995;LaPlaca and Thibault, 1998).…”

“…A similar mechanism was observed in PC12 cells. 29,30 Depending on the status of differentiation the amount of microtubules and microfilaments in PC12 was altered.…”
