Cytosine methylation affects the mutability of neighbouring nucleotides in human, Arabidopsis, and rice

Abstract: Methylated cytosines deaminate at higher rates than unmethylated cytosines and the lesions they produce are repaired less efficiently. As a result, methylated cytosines are mutational hotspots. Here, combining rare polymorphism and base-resolution methylation data in humans, Arabidopsis thaliana, and rice (Oryza sativa), we present evidence that methylation state affects mutation dynamics not only at the focal cytosine but also at neighbouring nucleotides. In humans, contrary to prior suggestions, we find that… Show more

“…In another investigation it was reported that methylated cytosine in human, Arabidopsis, and rice is not intrinsically associated with higher mutation risk for surrounding bases, but that it affects the mutability of neighboring nucleotides. [41] In an experimental investigation of direct kinetic measurements by LC-MS/MS sequencing study examining the influence of 5MeC on kinetics of p53 gene abduction by benzo[a]pyrene-7,8- dihydrodiol-9.10-epoxide (BPDE) it was reported that 5MeCÀ G sites enhances the reactivity of DNA towards electrophiles. [42] In an earlier investigation employing circular dichroism and UV-vis spectroscopy it was reported that cytosine methylation in a CpG sequence leads to enhanced reactivity with BPDE and it induces structural changes in duplex DNA by slightly unwind the DNA helix.…”
