Cytoprotective Effect of Heat Shock Protein 27 Against Lipopolysaccharide-Induced Apoptosis of Renal Epithelial HK-2 Cells

Abstract: Background:In response to various stimuli, heat shock protein 27 (Hsp27) functions as an anti-apoptotic or/and anti-inflammatory factor which confers a survival advantage to cells. This study was aimed to explore whether Hsp27 also has a cytoprotective role in human renal tubular epithelial cells, and to evaluate its potential in treating septic acute kidney injury (septic AKI). Methods: HK-2 cells were subjected to different concentrations (0-10 μg/mL) of lipopolysaccharide (LPS) for various times (0-24 h) to… Show more

“…To further explore the mechanism of lncRNA TapSAKI in sepsis‐induced kidney injury, PTEN and TLR4/NF‐κB signaling pathway were focused in LPS‐treated HK‐2 cells. LPS is the component of the cell wall of Gram‐negative bacteria, and studies have proved it is closely related to renal tubular epithelial cell apoptosis . Besides, the apoptosis of renal tubular epithelial cell is considered as an important cause of septic acute kidney injury .…”

[4,20] Besides, the apoptosis of renal tubular epithelial cell is considered as an important cause of septic acute kidney injury. [21] So, we considered HK-2 cell apoptosis as an indicator for septic acute kidney injury.…”

“…The concentration of Ang II or LPS was selected according to previous studies. In Ang II-induced inflammation model, HK-2 cells were treated with 1 μ M of Ang II or PBS for 12 h. 44 , 46 In LPS-induced inflammation model, HK-2 cells were treated with 5 μ g/ml of LPS or PBS for 8 h. 47 , 48 …”
