Cytoprotection-organoprotection by somatostatin: gastric and hepatic lesions

Abstract: In rats, the hemorrhagic gastric erosions produced by ethanol, and the fatal hemorrhagic hepatic necrosis induced by phalloidin, were significantly reduced by regular somatostatin, but not by derivatives devoid of -SH containing cysteines. These effects of the hormone were abolished in animals which received, in addition, the sulfhydryl blocker n-ethylmaleimide before the toxic chemicals. Thus, somatostatin exhibits organoprotection dependent on endogenous sulfhydryls.

“…The effects of somatostatin and octreotide on acute pancreatitis have been attributed to inhibition of the exocrine pancreatic functions [22, 23]. Increased hepatic and splenic reticuloendothelial system activity and modulation of the immune and cytokine responses have been noted following octreotide treatment [24], and may represent another mechanism of action, presumably a ‘cytoprotective effect’ [25, 26]. …”

“…This effect of somatostatin may be beneficial in both preventing early recurrent bleeding from gastric ulcers and in promoting ulcer healing. There is also evidence from experimental studies that somatostatin activates blood coagulation [56] and that it is cytoprotective to the stomach and liver [57,58]. Somatostatin has also been reported to cause a rapid significant increase (26 vs. 34 mm Hg; p !…”
