Cytoplasmic IκBα Increases NF-κB-independent Transcription through Binding to Histone Deacetylase (HDAC) 1 and HDAC3

Abstract: IB␣ is an inhibitory molecule that sequesters NF-B dimers in the cytoplasm of unstimulated cells. Upon stimulation, NF-B moves to the nucleus and induces the expression of a variety of genes including IB␣. This newly synthesized IB␣ also translocates to the nucleus, removes activated NF-B from its target genes, and brings it back to the cytoplasm to terminate the phase of NF-B activation. We show here that IB␣ enhances the transactivation potential of several homeodomain-containing proteins such as HOXB7 and P… Show more

“…Immunoprecipitations, immunofluorescences and isolation of enriched native chromatin extracts Immunoprecipitations in 293 cells were carried out as previously described (Viatour et al, 2003). For immunofluorescence studies, HeLa cells were seeded in six-well plates and subsequently transfected with the indicated expression plasmids using FUGENE 6 (Roche).…”

“…Use of dominant-negative IKK or IB-SR does not allow for the determination of a potential role of specific NF-B subunits in the transformation process. Additionally, overexpression of IB␣ or IB-SR can have effects on cells that are independent of NF-B (22,23). Therefore, to determine whether NF-B subunits, specifically p65 and c-Rel, have essential roles in Ras-induced transformation, we used genetic knockout-derived MEFs.…”

“…Interestingly, the motif for POU1F1 (also known as PIT-1) is significantly enriched in the promoters regulating transcripts that are downregulated early during NOD2 activation. The transactivation potential of this homeodomain-containing protein is enhanced via IkBa through an NF-kB-independent association with histone deacetylase 1 and 3 (39).…”
