Cytokine secretion by human fetal membranes, decidua and placenta at term

Abstract: Cytokines such as monocyte chemotactic peptide-1 (MCP-1), interleukin-8 (IL-8), RANTES (Regulated on Activation and Normally T-cells Expressed and presumably Secreted) and interleukin-10 (IL-10) are thought to play pivotal roles in immune recognition, acceptance of the fetal allograft, maintenance of pregnancy and parturition. Their secretion and regulation within the third trimester uterus is, however, less well defined. We therefore investigated the release of these cytokines by third trimester amnion, chori… Show more

“…During labour there is increased expression of proinflammatory mediators. These include pro-inflammatory cytokines, such as IL1b, IL6 and IL8 in the cervix, myometrium, foetal membranes and placenta (Denison et al 1998, Young et al 2002, Osman et al 2003; prostaglandins PGE 2 and PGF 2a in the foetal membranes and myometrium (Olson 2003); and matrix metalloproteinase 9 (MMP9) in the placenta and foetal membranes (Xu et al 2002). Regulation of these mediators facilitates tissue remodelling of the cervix, myometrial contractions and rupture of membranes, leading to successful delivery at term.…”

“…Because RANTES is produced in human placental tissues (Denison et al, 1998) and can induce the migration of leukocytes and other cell types (Youngs et al, 1997), we speculated that this chemokine is a candidate for a DPPIV substrate that can affect trophoblastic migration. To substantiate this possibility, we initially examined the expression of RANTES receptors on trophoblasts.…”

“…Several chemokines, including CCR1 ligands, have been demonstrated in human placental tissue (Denison et al, 1998;Drake et al, 2001;Red-Horse et al, 2001). They are considered to be involved in either recruiting specific leukocyte populations to the feto-maternal interface or modulating trophoblastic function (Ishii et al, 2000).…”

“…Of particular interest is the chorionic membrane; it is the largest area of contact with the maternal tissues and it is the first tissue colonized by microbial pathogens during an ascending intrauterine infection. In vitro, human choriodecidua produces a 10-fold higher level of the proinflammatory cytokines TNF-␣ and MCP-1 than human amnion (7,8).…”
