Cytokine- or chemically derived nitric oxide alters the expression of proteins detected by two-dimensional gel electrophoresis in neonatal rat islets of Langerhans.

Abstract: Interleukin-1␤ (IL-1␤) treatment of neonatal rat islets19 of a total of 1,600 detectable proteins in GSNOtreated islets (P < 0.01). We conclude 1) that the expression of up to 42 proteins is altered by cytokineinduced or chemically generated NO in the precise experimental conditions chosen and 2) that the majority of proteins altered by prior treatment with IL-1␤ may be the result of NO-independent IL-1␤-mediated regulation of gene expression. This study demonstrates that the combination of 2D gel electrophore… Show more

“…NO modifies the expression of several genes and proteins in beta cells [10,44,45]. The addition of the iNOS inhibitor L-MA prevented cytokine-induced NO production by the human islets, but did not affect the induction of mRNAs encoding IL-15 and the chemokines analyzed (Fig.…”

“…protein expression (John et al 2000, Mose Larsen et al 2001) of 'harmful' and 'protective' proteins in -cells. Our work has shown that a previously unreported potentially harmful action of cytokines is inhibition of an existing antioxidant defence.…”

“…We have shown previously that IL-1β induces PIPP in islets [8,9,10]. PIPPs comprises up-regulation and down-regulation of protein expression and/or gelposition-shifts resulting from post-translatory modifications.…”

“…haeme oxygenases and manganese superoxide dismutase (MnSOD)] [5,6]. Many IL-1β effects are mediated by inducing the inducible nitric oxide syntase (iNOS) and its product, nitric oxide [7]; however, NO-independent mechanisms also occur [8,9,10]. We hypothesize that the beta cell, when exposed to IL-1β initiates a protective response in competition with a series of deleterious events, and that in beta cells the deleterious events prevail [3].…”
