Cytokine and Chemokine Responses Associated with Clearance of a Primary Salmonella enterica Serovar Typhimurium Infection in the Chicken and in Protective Immunity to Rechallenge

Abstract: Infection of poultry with Salmonella enterica serovar Typhimurium poses a significant risk to public health through contamination of meat from infected animals. Vaccination has been proposed to control infections in chickens. However, the vaccines are currently largely empirical, and our understanding of the mechanisms that underpin immune clearance and protection in avian salmonellosis is not complete. In this study we describe the cytokine, chemokine, and antibody responses and cellular changes in primary an… Show more

“…It was also shown that after infection with Salmonella the number of CD8 + TCRγδ + T-cells in the chicken intestine increased [6]. CD4 + or CD8 + T-cell responses are not required for the early host response [14], but CD4 + T-cell responses, particularly Th1 responses, play an important role in the clearance of Salmonella from the gastrointestinal tract [5,10,27].…”

“…LITAF is required throughout Salmonella infection. IL-12, secreted by activated macrophages, acts both independently and synergistically on natural killer cells and T helper (Th) cells to induce the production of further IFN-γ, which activates the macrophages through a positive feedback loop (Beal et al, 2004;Withanage et al, 2005;Haghighi et al, 2008;Tohidi et al, 2012). A correlation between cytokine profiles of chickens and their resistance to the infection by Salmonella Typhimurium was found (Mead, 2000).…”

“…Withanage et al (2005) reported that the expression of IL-6, well-known pro-inflammatory cytokine, was increased in the caecal tonsils, ileum, and spleen on day 1 p.i. and day 3 p.i.…”

“…The Guildhay method requires an initial serum dilution of 1:500 whereas similar work with killed Typhimurium used initial serum dilution of 1:250, 1:400 and 1:400 respectively Withanage et al, 2005;Bailey et al, 2007a). The higher initial dilution rate may have greatly reduced the number of ELISA positives in this trial.…”
