Cytogenetic mapping of 31 functional genes on chicken chromosomes by direct R-banding FISH

Abstract: Using direct R-banding fluorescence in situ hybridization, we determined the location of 31 functional genes on chicken chromosomes. Replication R-banded chromosomes were obtained by synchronizing splenocyte cultures with excessive thymidine, followed by BrdU treatment. Thirty-one functional genes were directly localized to banded chicken chromosomes using genomic DNA and cDNA fragments as probes. The possibility of conserved linkage homology between chicken and human chromosomes was demonstrated for seven chi… Show more

“…The karyotypic similarities and differences between bird species have been studied morphologically by conventional Giemsa staining and chromosome banding, and, for the past 10 years, molecular cytogenetically by comparative FISH mapping with chromosome-specific paints, cDNA and genomic DNA clones, mostly developed in chicken (Gallus gallus) , Suzuki et al 1999, Schmid et al 2000, Shibusawa et al 2001. Cross-species chromosome hybridization (termed Zoo-FISH) and subsequent comparative gene mapping delineates accurately the chromosomal orthologies between distantly related species and the chromosome rearrangements that have occurred during evolution.…”

“…Five genes (TH, INS, CCND1, PAX6, RAG2) that lie in the p15-q13 region of human chromosome 11 have been mapped to this region of chicken chromosome 5 by in situ hybridization [23,24]. Whereas TH, INS, and CCND1 have been mapped to 11p15 and to mouse chromosome 7, PAX6 and RAG2 lie in 11p13 and on mouse chromosome 2 (Mouse Genome Database, 2001).…”
