Cytodiagnosis of Malignant Melanoma: Immunoperoxidase Staining with HMB-45 Antibody as an Aid to Diagnosis

Abstract: The specificity and sensitivity of HMB-45, an antimelanoma monoclonal antibody, was evaluated in cytologic specimens from extracutaneous melanomas. Melanoma cells in 23 of 25 (92%) cases stained with this antibody. Staining was intense and diffuse in 22 of these melanomas and focal in 1. Amelanotic tumors and tumors with scanty pigment were included. Neither degree of pigmentation, site of primary, cytologic characteristics of tumor, nor source of specimen predicted the immunostaining pattern. Cytopreparations… Show more

“…4 Thanks to its high specificity, this antibody also appeared to be a useful marker for malignant melanoma cells in smears prepared from cytologic specimen. 15 HMB-45 immunoreactivity has also been reported in a rare variant of signet ring cell melanoma lacking a well-known marker of melanoma such as S-100 protein. 2 In this study we have demonstrated that 2% of breast carcinomas show HMB-45 immunoreactivity.…”

“…To analyze gene transfer efficiency, gp100 expression of DC was determined by immunohistochemistry staining with anti-gp100 mAb HMB-45 as previously described. 36 Briefly, cells were fixed and blocked and incubated for 1 h with the HMB-45 mAb (DAKO-HMB45, M634, Hamburg, Germany). Ag expression was detected by a second step Ab (DAKO) followed by ABC staining (DAKO).…”

“…Special stains that are relatively specific for melanoma, such as S-100 9 or HMB-45, 10 have been available for years, but their use has never been incorporated into the routine pathological examination of lymph node specimens. This is mainly because immunohistochemical analysis of all the nodes from a complete lymph node dissection, which typically involves 10 to 30nodes,wouldbeprohibitivelytime-consuming and expensive.…”
