Cytocompatibility of Ti–6Al–7Nb through High-Pressure Torsion Processing

Abstract: In this study, we evaluated the cytocompatibility of high-pressure torsion (HPT)-processed Ti-6Al-7Nb alloys. Grain size of titanium (Ti) alloys decreased from 5 µm (without HPT processing, HPT-0) to 100 nm (at 2 GPa, HPT-2) or 70 nm (at 6 GPa, HPT-6) through HPT processing. Cell adhesion, proliferation, and differentiation were evaluated with mouse preosteoblast (MC3T3-E1). A locomotion trend was presented by cells cultured on HPT-2 to compare with other specimens, while, an immobilization trend was presented… Show more

“…The adhesion and differentiation of hMSCs cultured on all specimens were quantitatively evaluated by real-time reverse transcriptase-polymerase chain reaction (RT-PCR) analysis of selected target genes (Table I, synthesized by Greiner Bio-One, Tokyo, Japan) following a previously published protocol. 27 Total RNA from each specimen was extracted using an RNeasy Kit (Qiagen GmbH, Hilden, Germany) according to the manufacturer's protocol, and 2 μg of RNA was reverse-transcribed to first-strand cDNA using Pri-meScript II first Strand cDNA Synthesis system (Takara Biotechnology Co., Dalian, China) according to the manufacturer's instructions. The reaction cocktail for realtime RT-PCR contained 10 μL of SYBR Premix Ex Taq II (Takara Biotechnology Co.), each forward and reverse primers at 10 pmol, 2 μL of cDNA, and distilled water to a final volume of 20 μL.…”

“…Two target genes involved in the initial osteogenic differentiation activation of preosteoblasts and two additional target genes involved in osteogenic differentiation and calcification were selected and validated (Table ). The expression of the four target genes was determined by following a previously published protocol . Total RNA was extracted from the cells cultured on fsTi and mTi using an RNeasy Kit (Qiagen GmbH, Hilden, Germany) in accordance with the manufacturer's instructions.…”

“…The expression of the four target genes was determined by following a previously published protocol. 19,20 Total RNA was extracted from the cells cultured on fsTi and mTi using an RNeasy Kit (Qiagen GmbH, Hilden, Germany) in accordance with the manufacturer's instructions. Two micrograms of RNA were reverse-transcribed into firststrand complementary DNA with the PrimeScript II 1st Strand cDNA Synthesis Kit (Takara Biotechnology Co. Ltd., Dalian, China), which uses random hexamers, in accordance with the manufacturer's protocol.…”

“…By contrast, cells growing on the CCM HPT specimen and the CCM HPTA specimen display a pointed distribution of adhesion plaques in cells. As known the role of vinculin during cell adhesion behavior, 20,21) the shapes of cells cultured on the CCM HPT specimen indicate a stronger cell locomotion trend, on the other hand, those on the CCM ST specimen present a stronger immobilization trend. Cells cultured on the CCM HPTA specimen show an intermediate pattern.…”
