Cytochrome P4501A1, glutathione S-transferase M1 and T1 gene polymorphisms in chronic myeloid leukemia

Role of Glutathione-S-Transferase M1 (GSTM1) and T1 (GSTT1) Genes in the Development and Progress of Chronic Myeloid Leukemia and in the Formation of Response to Imatinib Therapy

“…Genotype GSTT1*00 was 2.67 times more incident in the patients than in controls (35.0 and 13.1%, respectively,  2 =23.62; p<0.0001). Homozygotic carriership of GSTT1*00 "zero" allele increased the CML risk 3.66 times vs. GSTT1*+ carriership (OR=3.66, 95% CI=2.13-6.30), which was almost 1.8 times higher than the value estimated in the previous report [2]. The data on the pathogenetic signifi cance of these genes polymorphisms in CML are scanty and contradictory [2,10,11].We studied the GSTM1 and GSTT1 "zero" polymorphisms and their combinations as probable factors essential for the risk of CML development and progress and for a high cytogenetic response after 12-month imatinib monotherapy in a daily dose of 400 mg in the Russians living in the Vyatka region.
