Cysteinyl Leukotrienes Promote Human Airway Smooth Muscle Migration

Abstract: Cysteinyl leukotrienes promote airway smooth muscle (ASM) contraction and proliferation. Little is known about their role in ASM migration. We investigated this using cultured human ASMs (between the second and fifth passages) obtained from the large airways of resected nonasthmatic lung. Platelet-derived growth factor-BB (1 ng/ml) promoted significant (3.5-fold) ASM migration of myocytes across collagen-coated 8- micro m polycarbonate membranes in Transwell culture plates. Leukotriene E(4) (10(-7), 10(-8), 10… Show more

“…CYSLTR1 encodes the G-proteincoupled receptor CysLT 1 . Cys-LTs are biologically active lipid mediators, which can promote human airway smooth muscle contraction, proliferation and migration [29]. Cys-LTs have also been reported to be involved in the generation of type-2 T-helper cell cytokines and expression of some mediators involved in the process of airway inflammation and remodelling, such as TNF-a, endothelin 1 [30], reactive oxygen intermediate [31], histamine receptor [32] and IL-11 [33].…”

“…It revealed that genes related to extracellular matrix, transcription, and cellular stress responses were deregulated in fibroids. Collagen has been shown to directly stimulate smooth muscle cell growth (Herz et al, 2003) and regulate cell migration (Parameswaran et al, 2004). Increased expression of collagens and other extracellular matrix proteins could thus have an important role in fibroid growth.…”

“…Migration is required for each of these alternative mechanisms for smooth muscle hyperplasia. Migration of ASM can be induced by plateletderived growth factor [116,117], enhanced by cys-leukotrienes [118] modulated by b 2 -adrenoceptor agonists and glucocorticoids [119], with the latter agents losing activity when cells are seeded onto a matrix containing monomeric type-1 collagen [120,121].…”
