CYP3A activity in African American and European American men: Population differences and functional effect of the CYP3A4*1B 5′-promoter region polymorphism

Abstract: Comparison of CYP3A activity between populations is complicated by frequency distribution differences in the regulatory CYP3A4*1B polymorphism and lower hepatic CYP3A activity associated with the variant allele. However, this reduction is modest; therefore no major and clinically important difference in CYP3A activity is present between Americans of African or European descent.

“…In the present study, CYP3A4*1B decreased the inducible thiotepa clearance. Other authors have shown that CYP3A4*1B caused a modest decrease in systemic midazolam [38] and ciclosporin clearance [39,40], whereas others did not find an effect [41,42].…”

“…12 In addition, black men are more likely to possess genetic polymorphisms that favor increased androgen activity such as shorter CAG tri-nucleotide repeats in the androgen receptor gene 13 and a possibly less active variant of the CYP3A4 gene, which encodes a protein involved in the oxidative deactivation of testosterone. 14 Finally, increased androgen activity among black men was suggested by a study which found that benign and malignant prostate tissue from black men had both a greater number of cells staining positive for the androgen receptor as well as more intense staining within positive cells. 15 Although androgens regulate cellular PSA production they also affect prostate size.…”

“…Several studies assessed the risk of SCCHN in relation to CYP or GST genotypes (20 -22). However, interindividual variations in the expression and activity of the CYPs and GST are dependent on genotypic as well as post-transcriptional factors, which maybe tissue specific (23)(24)(25). Therefore, the most direct method to evaluate the effects of CYPs and GST on the carcinogenic effects of tobacco is to evaluate enzyme expression in the tissue of interest.…”
