Cylindrotheca is one of the main model diatoms for ecophysiological and silicification research and is among the few diatoms for which a transformation protocol is available. Knowledge of its life cycle is not available, however, although sexual reproduction has been described for several related genera. In this study, 16 Cylindrotheca closterium strains from a single rbcL lineage were used to describe the life cycle of this marine diatom, including the sexual process and mating system. Similar to other Bacillariaceae, sexual reproduction was induced by the presence of a suitable mating partner, with two gametes produced per gametangium, resulting in two auxospores per gametangial pair. Differences with other Bacillariaceae include details of cell pairing, gamete behaviour, auxospore orientation and chloroplast configuration, and perizonium structure. The mating system is heterothallic, since strains fell into two mating type groups, with several strains of one mating type occasionally displaying intraclonal auxosporulation. Initial cell lengths were on average 95-100 µm, the sexual size threshold was at least 66 µm, and the minimal viable cell length c. 11 µm. Sexual reproduction could be synchronized by dark conditions, which allowed us to determine that the whole sexual process is completed in less than 24 h. Furthermore, large percentages of cells at defined sexual stages can easily be obtained and the possibility to experimentally manipulate the life cycle provides a valuable tool for future research on all aspects of the biology of this model diatom.

Knowledge on benthic diatom taxonomy and hence biodiversity from Arctic locations is rare, though the few existing analyses address morphological characteristics only. Morphological treatment is a time-consuming process and just allows the taxonomic identification of a low sample volume. In contrast, molecular sequencing is a faster and universally applicable approach, and also has the potential for refined analyses. Unfortunately, a lot of molecular information in databases is not trustable, and often existing genetic data do not fit together with morphological aspects. Therefore, the main goal of our study was for the first time to combine both morphological features and DNA information on benthic Arctic diatoms. In a first step, we studied the morphology of 26 strains from the Rostock Culture Collection of Arctic diatoms via light and electron microscopies. Secondly, we addressed their molecular taxonomy by sequencing the DNA barcode 18S V4 and the chloroplast marker rbcL. As a result, we could identify five species. Seven of the altogether twelve taxa could not be attributed to any known species, because listed sequences and morphology did not fit together. From these, three species were described as new to science here. Although the studied strains represent only a minor fraction of the diatom flora of Svalbard, this study is a first milestone in coupling morphological and molecular data of benthic diatoms in such high latitudes. By making these data available online, they contribute to a taxonomic reference library for Arctic benthic diatoms and are available for comparative studies.

Many diatom and other microbial eukaryote morphospecies consist of a variable number of (pseudo)cryptic species, with obvious consequences for such fields as biogeography and community ecology. Here, we investigated the species limits of morphologically similar small-celled strains of the model diatom Sellaphora from the United Kingdom and Australia, using cox1 mitochondrial and rbcL chloroplast gene sequences. Based on cox1 sequence data, the sequenced strains belonged to six closely related lineages, presumably species, of which one corresponds to the previously described S. auldreekie D.g. mann & s.m. mCDonalD. Although rbcL displayed less sequence variation, the same six lineages were also recovered in an rbcL phylogeny of the genus. Molecular species discovery was compared to mating trials involving three of the lineages, showing that they were reproductively isolated. Incomplete evidence from a fourth lineage suggested that it too was reproductively isolated. A posteriori examination of light microscope morphology revealed no simple metrics or presence/absence characters that could consistently separate all species of the auldreekie complex, even though some do differ in pole width or stria density. While it is premature to make conclusions about their biogeography, it is obvious that a number of cryptic Sellaphora species thus far undetected in the UK are easily found at several localities in warm-temperate Australia.
