Cyclosporine A loaded brain targeting nanoparticle to treat cerebral ischemia/reperfusion injury in mice

Abstract: Background Ischemic stroke is one of the main causes of death and disability in the world. The treatment for ischemic stroke is to restore blood perfusion as soon as possible. However, when ischemic brain tissue is re-perfused by blood, the mitochondrial permeability transition pore (mPTP) in neuron and microglia is excessively opened, resulting in the apoptosis of neuron and nerve inflammation. This aggravates nerve injury. Cyclosporine A (CsA) inhibits the over-opening of mPTP, subsequently r… Show more

“…32 Previous findings have shown that CsA is capable of preventing ischemic brain injury by inhibiting the massive activation of microglia and the production of cytokines in rats with chronic cerebral hypoperfusion. 33 Microglia are of great significance in central sensitization and the development of NP. Abnormally activated microglia following peripheral nerve injury causes abnormal excitation of spinal dorsal horn neurons by releasing neuromodulators.…”

“…In contrast, the neuroprotective cytokine IL-10 was significantly increased. [259] Poly(dopamine) NPs (PDA NPs), as the name implies, are nanostructures obtained from the polymerization of dopamine into poly(dopamine), an artificial melanin-like material. [260] PDA NPs are a promising polymeric-like candidate for treating CNS diseases, as they possess antioxidant properties attributed to their abundant phenolic hydroxyl groups.…”
