Cyclophosphamide loaded albumin microspheres II. Release characteristics

Abstract: The purpose of this investigation was to evaluate the release characteristics of cyclophosphamide (CP) from glutaraldehyde stabilized human serum albumin microspheres, and to study the effect of the extent of cross-linking, the amount of the stabilizing agent and the size of the microspheres on the in vitro release of CP. Microspheres were prepared by emulsion polymerization method using two different volumes (0.1 and 0.7 ml) of glutaraldehyde solution (25 per cent) and two different crosslinking durations (15… Show more

“…Albumin nanoparticles have been extensively investigated with respect to their preparation methods and release properties Loo et al, 2004;Vural, Kas, Hincal, & Cave, 1990). Human serum albumin (HSA) and bovine serum albumin (BSA) have been used as natural matrix materials for delivery devices (Brannon-Peppas, 1995).…”

“…A burst initial phase in the release profiles of drugs encapsulated in Alb MS has been reported by various authors (Gupta et al, 1986;Vural et al, 1990;Mathew et al, 2007;Gülsu et al, 2012) and has been attributed to fast dissolution of loosely bound drug located near the surface followed by prolonged release of the remaining drug payload encapsulated deeper within the MP.…”

“…Accordingly only 25% drug theoretical loading generated MS with the highest EE%. A commonly used crosslinker, glutaraldehyde, might increase drug EE with/without affecting the PS (Vural et al, 1990). Its long-term use has been associated with toxicity issues so it is important to use it at an optimal minimum concentration to avoid potential health problems (Gülsu et al, 2012).…”

“…A large number of papers published in this area can be exemplified by those on streptomycin sulfate (Gurkan et al, 1987)-, rifampicin (Igeri et al, 1989(Igeri et al, , 1991-, cyclophosphamide (Vural et al, 1990(Vural et al, , 1991)-, and chloramphenicol (Onur et al, 1993)-loaded gelatin and/or albumin microspheres, and furosemide-containing chitosan microspheres (Akbuga and Durmaz, 1994). Giirsel and Hasirci (in press) constructed microcapsules from poly (3-hydroxybutyric acid) and its copolymers and polyphosphate-Ca2+ complex.…”
