Cyclophilin C-Associated Protein and Cyclophilin C mRNA are Upregulated in Penumbral Neurons and Microglia after Focal Cerebral Ischemia

Abstract: Immunophilin ligands, such as cyclosporin A and FK506, have neuroprotective effects in experimental stroke models, although the precise mechanism is unclear. Cyclophilin C-associated protein (CyCAP) is a natural cellular ligand for the immunophilin, cyclophilin C, and has a protective effect against endotoxins by downmodulating the proinflammatory response. Expressions of CyCAP and cyclophilin C mRNA in a rat middle cerebral artery (MCA) occlusion ischemia model were investigated by Northern blotting and in si… Show more

“…Cyclophilin C-associated protein ( CyCAP ) is a 77 kDa intracellular protein, with a scavenger-receptor cysteine-rich domain that is released into the cytoplasm and cell membrane [23,24,25]. The expression of CyCAP has been shown to be elevated by wounds and ischemia due to the presence of interferon-gamma (IFNγ) and tumor necrosis factor-alpha ( TNFα ) [26,27]. In addition, the deletion of CyCAP abolished a 45 kDa fibronectin-induced MMP-13 expression [28,29].…”

“…Recently, evidence indicates that the lectin family is involved in protein folding and sorting in the secretory pathway and ER (Schrag et al, 2003). The most intensively studied ER lectin chaperones are calnexin and calreticulin (Saito et al, 1999; Danilczyk et al, 2000; Snapp et al, 2006; Williams, 2006). Both calnexin and calreticulin can bind newly synthesized glycoprotein and participate in their correct folding before the protein is secreted.…”

“…Inflammation is mediated by both molecular components, including cytokines, and cellular components, mainly microglia, many of which have pro-and/or antiinflammatory properties [40]. Increased levels of two glial proteins related to the inflammatory response have been observed in Alzheimer's transgenic mice: glial fibrillary acidic protein (GFAP), which overexpression was confirmed by western blotting, and the complement protein C1q.…”
