Cyclophilin 20-3 relays a 12-oxo-phytodienoic acid signal during stress responsive regulation of cellular redox homeostasis

Abstract: The jasmonate family of phytohormones plays central roles in plant development and stress acclimation. However, the architecture of their signaling circuits remains largely unknown. Here we describe a jasmonate family binding protein, cyclophilin 20-3 (CYP20-3), which regulates stress-responsive cellular redox homeostasis. (+)-12-oxo-phytodienoic acid (OPDA) binding promotes CYP20-3 to form a complex with serine acetyltransferase 1, which triggers the formation of a hetero-oligomeric cysteine synthase complex … Show more

“…CYP have been found to be involved in the signalling of several phytohormones, including auxin, brassinosteroid, gibberellic acid (GA) and jasmonic acid 16,17,[23][24][25][41][42][43] . Among these and other phytohormones, the activation of the signalling pathways of GA, jasmonic acid and strigolactone also utilize a derepression mechanism by the proteasomal degradation of specific repressors in a manner similar to that of auxin [44][45][46][47][48] .…”

“…Preferential expression of genes upon OPDA treatment such as ZAT10 or GRX480 together with about 150 genes have been postulated repeatedly, 18,26,28 and also proteomic approaches showed OPDA-specific responses. 27 In all these experiments, however, putative formation of JA upon OPDA treatment was not excluded.…”

“…26 GRX480 encodes a GLUTAREDOXIN and is regarded as candidate for an OPDAassociated transcription regulator. 18 Both genes were previously shown to be OPDA-inducible, 18,26 indicating their role as a putative marker for OPDA responses. COI1-independent expression was confirmed for GRX480, since tests with the coi1-16 mutant showed an OPDA-inducible expression of GRX480.…”
