Cyclooxygenase-2 Unlike Cyclooxygenase-1 is Highly Expressed in Ovine Embryos during the Implantation Period1

Abstract: In this study we investigated expression of the two isoforms of the prostaglandin-forming enzyme, cyclooxygenase-1 (Cox-1) and cyclooxygenase-2 (Cox-2), in sheep embryos. Using Western blot and immunohistochemical analyses, we demonstrated that Cox-2 was highly expressed in embryos from Day 8 to Day 17 of development whereas Cox-1 was undetectable during this time. The expression of Cox-2 was developmentally regulated. It was maximal between Days 14 and 16. There was a 30-fold increase in Cox-2 content per pro… Show more

“…The termination of PTGS2 expression on the embryo coincides with implantation in sheep (Charpigny et al 1997). Consistent with our data of no further rise after days 15-18, day 19 bovine blastocysts secreted PG less abundantly than those of day 16 (Lewis et al 1982), obviously peaking around day 18 (Wilson et al 1992) depending on the specific PG in focus.…”

“…PTGS2 was demonstrated in trophoblast cells (Charpigny et al 1997), and the supernatant of blastocysts contained considerable amount of PG in cattle and sheep (Marcus 1981, Charpigny et al 1997.…”

“…Prostaglandins have an important role in the regulation of the estrous cycle and successful implantation in pigs (Spencer and Bazer 2004). PTGS and PTGS2 are responsible for the formation of prostanoids, including prostaglandins (Sawdy et al 2000;Charpigny et al 1997). In our study, we noted the upregulation of PTGS2 caused by adiponectin treatment.…”

“…Expression of PTGS2 is more abundant in blastocysts resulting in calf delivery compared with those resulting in resorption (El-Sayed et al 2006). In bovine, the PTGS2 protein localizes to the TE, while the ICM does not express PTGS2 (Charpigny et al 1997), suggesting that this gene is necessary for the elongation and subsequent implantation. Moreover, higher expression of PTGS2 throughout the implantation window indicates an important role for the PGs released by the embryo in mediating interactions with the uterus (Charpigny et al 1997, Wang et al 2002.…”

As PTGS2 expression is time regulated during ruminant embryo development, differential expression could be related to asynchronous development between IVF and parthenogenetic embryos.…”
