CX3CL1 (Fractalkine) Protein Expression in Normal and Degenerating Mouse Retina: In Vivo Studies

Abstract: We aimed to investigate fractalkine (CX3CL1) protein expression in wild type (wt) retina and its alterations during retinal degeneration in mouse model (rd10) of retinitis pigmentosa. Forms of retinal protein CX3CL1, total protein and mRNA levels of CX3CL1 were analyzed at postnatal days (P) 5, 10, 14, 22, 30, 45, and 60 by Western blotting and real-time PCR. Cellular sources of CX3CL1 were investigated by in situ hybridization histochemistry (ISH) and using transgenic (CX3CL1cherry) mice. The immunoblots reve… Show more

“…In support of this, fractalkine cleavage increases prior to neuronal cell death in vitro [71] and loss of membrane-bound fractalkine precedes degeneration in the rd10 retina [50]. Western blots of P20 retinas revealed an increase in full-length (Fig 8C; 100kDa) and a decrease in soluble (Fig 8C; 85kDa) isoforms of fractalkine with Norgestrel.…”

“…Membrane-bound fractalkine is cleaved by metalloproteinases, such as ADAM10, to release soluble fractalkine (85kDa) [53]. Fractalkine is also recycled from the membrane and stored in intracellular vesicles [51, 54], possibly as a 95kDa isoform [50]. Signaling to its receptor CX3CR1 on microglia [55], both in vivo and in vitro studies have shown that complementary fractalkine-CX3CR1 signaling can modulate microglial activation and migration resulting in improved neuronal survival [5661].…”

“…CX3CL1 is expressed on the surface neurons in the CNS and endothelial cells in non-neuronal tissues [22]. In the mouse retina, CX3CL1 is expressed on ganglion cells and within the inner plexiform layer [23]. Activation and downstream effects of the CX3CL1-CX3CR1 signaling are complex, tissue-dependent and are likely influenced by many factors.…”

“…Since the FKN ligand is enriched in the ganglion cell layer of the retina in both humans and mice (Silverman et al, 2003; Zieger et al, 2014) including RGCs in the DBA/2J (Supplemental Figure 1), we reasoned that loss of FKN signaling may influence key aspects of RGC decline in glaucoma. Deficits in axonal transport progressively affect RGC axon and somal integrity in glaucoma (Soto et al, 2008; Crish et al, 2010; Dengler-Crish et al, 2014).…”
