Cutting Edge: Th17 and Regulatory T Cell Dynamics and the Regulation by IL-2 in the Tumor Microenvironment

Abstract: Th17 cells play an active role in inflammation and autoimmune diseases. However, the nature and regulation of Th17 in the context of tumor immunity remain unknown. In this study, we show that parallel to regulatory T (Treg) cells, IL-17+ CD4+ and CD8+ T cells are kinetically induced in multiple tumor microenvironments in mice and humans. Treg cells play a crucial role in tumor immune pathogenesis and temper immune therapeutic efficacy. IL-2 is crucial for the production and function of Treg cells. We now show … Show more

“…In Although prior studies have reported that CD4 1 or CD8 1 T cells were implicated in IL-17 production in tumor-bearing hosts [3,34,35], these reports have not examined gd T cells. In our experiments, we showed that whereas small numbers of CD4 1 T cells produced IL-17, there were much more IL-17-producing gd T cells in the transplanted tumor tissues ( Fig.…”

“…It has been demonstrated that IL-17 is expressed in tumor microenvironments [3][4][5]. The precise role of IL-17 during tumor development, however, remains unclear.…”

“…Most studies in both mouse and humans have focused upon CD8+ cytotoxic T cells that kill tumors directly and T H 1 CD4+ T cells (producing IL-2, and IFN-Îł) that contribute directly or indirectly to tumor killing [43]. While T H 1 remains the dominant T H subset in tumors within the memory T cell pool, another T cell subset has recently been found in both mouse and human tumors that may play a significant role in the pathogenesis of tumors [44][45][46]. This subset is a CD4+ T cell that secretes IL-17A and has been labeled as the T H 17 cell [47,48].…”

“…In the presence of TGF-␀1 and IL-6 and absence of IL-2, substantially greater numbers of IL-17-producing cells are generated, as previously reported. 27,35 The addition of ATRA inhibited IL-17 expression in the absence of IL-2 while enhancing FoxP3 expression ( Figure 7A, lower right panel). We confirmed these results using IL-2-deficient Rag2 ÏȘ/ÏȘ TCRtransgenic (5C.C7) naive T cells ( Figure S1, available on the Blood website; see the Supplemental Materials link at the top of the online article).…”
