Curtobacterium flaccumfaciens pv. flaccumfaciens detection in bean seeds using a semi-selective culture medium

Abstract: The bacterial wilt caused by Curtobacterium flaccumfaciens pv. flaccumfaciens is currently considered one of the most important bacterial bean disease in Brazil. One of the most effective control methods against this disease is the use of healthy seeds. However, no methods are known that could be routinely used to detect this bacterium in bean seeds under Brazilian condition. The aim of this work was to evaluate qualitative and quantitative detection methods for Curtobacterium flaccumfaciens pv. flaccumfaciens… Show more

“…Detection of the pathogen on infected seeds is relatively easy due to their discoloration of yellow, orange or purple depending on the pigment of the bacterial strain (EPPO ). On the other hand, an effective semi‐selective culture medium has been developed for the detection of the bacterium in seed and leaf samples (Maringoni and Camara ; Camara et al. ).…”

“…Semi‐selective media for in vitro growth of C. flaccumfaciens pv flaccumfaciens are available (Mizuno and Kawai, ; Tegli et al., ; Maringoni and Camara, ; Maringoni et al., ), but methods based on isolation are quite time‐consuming and insufficiently specific (Tegli, ).…”

“…flaccumfaciens (Theodoro & Maringoni, 2006). O controle preventivo desta bactéria está fundamentado no uso de cultivares resistentes, rotação de culturas e uso de sementes sadias (Maringoni & Camara, 2006;Alencar et al, 2008;Herbes et al, 2008). Entretanto, o uso de variedades resistentes constitui um dos mais importantes meios de controle de doenças de plantas cultivadas, pois, além de ser uma das medidas mais econômi-cas, não traz risco de contaminação ambiental (Galli, 2005).…”
