Current Versus Lifetime Depression, APOE Variation, and Their Interaction on Cognitive Performance in Younger and Older Adults

Abstract: The absence of a depression by APOE interaction on cognitive abilities suggests that these synergistic effects only operate at the level of cognitive decline. This implies that it is those biological pathways especially affected by aging that become compromised further by the combined presence of depression and APOE e4 in an individual.

“…The association between self-rated memory and depressive symptom severity in older adults is well established (Kahn et al, 1975;Grut et al, 1993;Collins and Abeles, 1996;Jonker et al, 1996;Derouesne et al, 1999;Clarnette et al, 2001;Pearman and Storandt, 2004;Minett et al, 2005). Given the equally wellestablished link between depressive symptoms and cognition in older adults, (e.g., Burt et al, 1995;Koenig et al, 2015;Luciano et al, 2015;O'Shea et al, 2015), it is surprising that the association between subjective memory and objective memory performance is less clearly delineated in the literature. While some studies report an association (Gagnon et al, 1994;Jonker et al, 1996), other studies have not (Stewart et al, 2001;Jungwirth et al, 2004;Rickenbach et al, 2015).…”

“…With respect to the moderating effect of APOE E4 status, an important between-study variable is the measurement of key phenotypes. Studies that have used multiple cognitive tests and non-dichotomous depressive symptomatology measures have tended not to find the interaction (the present study, [ 22 , 23 ]; cf. [ 21 ]), whereas those that have used a cognitive impairment screener or clinical diagnosis of cognitive impairment along with a dichotimization or clinical diagnosis of depressive symptomatology have tended to find the interaction [ 12 18 ].…”

“…X chromosome polygenic scores were calculated in unrelated individuals from Generation Scotland [32] separately for men and women based on the sex-stratified association analyses. These were used to predict Eysenck Personality Questionnaire neuroticism scores, General Health Questionnaire 28-item [36] total scores (i.e., general psychological distress), and Structured Clinical Interview for DSM-IV (SCID)-diagnosed depressive disorder [37]. Following FDR correction, none of the results were significant (P > .05; Supplementary Table 5).…”
