Many people are affected by HIV infection worldwide, lack of ability of modern medication in developing an active vaccine against HIV is a global public health importance. Various aspects such as diseases associated with HIV infection, role of immune cells in diagnosis and treatment, significance of molecular changes during infection, complications in infants, impact of gender and social behaviour and diagnosis of disorders associated with HIV infection were discussed in this review. Affected patients would be associated with acute kidney injury, arterial stiffness, cardiovascular disease, Tuberculosis, intestinal parasite carriage and disruptive sleep apnea. Immune cells like CD4, CD8, CD38 T cells and natural killer cells have a vital role in diagnosis and treatment. DNA vaccines could play a major role to prevent or treat infectious diseases including HIV and would elicit T cell immunity, protective antibody responses and local mucosal immunity protective vaccine against HIV infection. Health professionals are responsible for diagnosis of disorders associated with HIV infection and follow-up of HIV-infected paediatric patients. There is a crucial need to improve and evaluate novel interventions that are tailored for this helpless population. Several factors such as income deficit, Pre-college residence, pornographic film viewing and believe in sexual abstinence for HIV prevention were found to be significant predictors of initiating premarital sex. Younger generations could be advised to maintain the virginity as cultural norm. Incorporation of socio-structural elements and geospatial techniques in analytical approaches would lead to better understanding of local dynamics and develop mediations that increase our facility of targeted HIV prevention services.

Genomics and molecular biology has always been a constant source of inspiration and motivational research for worldwide researchers in field of biology and biotechnology. These two fields have always generated a huge amount of data and in order to compile and analyze those, bioinformatics came into action during last decade. Implementation of bioinformatics has a clear intention of doing all these analysis of data in efficient and fast manner in order to cut down the expensive laboratory equipment, chemicals and most precious time. Mostly genomic data is composed of sequencing results at a higher scale and that is why manual curating and handling of these data is quite difficult. Supreme aim of this review is to make awareness about bioinformatics options in cancer genomics and viral genomics apart from next generation sequencing. Next generation sequencing or high throughput sequencing has helped a lot to replace old conventional method of sequencing and with the help of recent advances in technologies. These technologies are very efficient, fast and cheaper also as compare to conventional methods. Recently used methods were also discussed under appropriate headings. Role of bioinformatics is getting bigger and bigger in management and analysis of enormous amount of biological data generated through medical, biotechnological and clinical research globally. But we still need to understand challenges and limitation of bioinformatics as well as reliability.

Flow-through Capture and in Situ Amplification Can Enable Rapid Detection of a Few Single Molecules of Nucleic Acids from Several Milliliters of Solution

Detecting nucleic acids (NAs) at zeptomolar concentrations (few molecules per milliliter) currently requires expensive equipment and lengthy processing times to isolate and concentrate the NAs into a volume that is amenable to amplification processes, such as PCR or LAMP. Shortening the time required to concentrate NAs and integrating this procedure with amplification on-device would be invaluable to a number of analytical fields, including environmental monitoring and clinical diagnostics. Microfluidic point-of-care (POC) devices have been designed to address these needs, but they are not able to detect NAs present in zeptomolar concentrations in short time frames because they require slow flow rates and/or they are unable to handle milliliter-scale volumes. In this paper, we theoretically and experimentally investigate a flow-through capture membrane that solves this problem by capturing NAs with high sensitivity in a short time period, followed by direct detection via amplification. Theoretical predictions guided the choice of physical parameters for a chitosan-coated nylon membrane; these predictions can also be applied generally to other capture situations with different requirements. The membrane is also compatible with in situ amplification, which, by eliminating an elution step enables high sensitivity and will facilitate integration of this method into sample-to-answer detection devices. We tested a wide range of combinations of sample volumes and concentrations of DNA molecules using a capture membrane with a 2 mm radius. We show that for nucleic acid detection, this approach can concentrate and detect as few as ∼10 molecules of DNA with flow rates as high as 1 mL/min, handling samples as large as 50 mL. In a specific example, this method reliably concentrated and detected ∼25 molecules of DNA from 50 mL of sample.
