Current Status and Future Prospects of Genome-Scale Metabolic Modeling to Optimize the Use of Mesenchymal Stem Cells in Regenerative Medicine

Abstract: Mesenchymal stem cells are a promising source for externally grown tissue replacements and patient-specific immunomodulatory treatments. This promise has not yet been fulfilled in part due to production scaling issues and the need to maintain the correct phenotype after re-implantation. One aspect of extracorporeal growth that may be manipulated to optimize cell growth and differentiation is metabolism. The metabolism of MSCs changes during and in response to differentiation and immunomodulatory changes. MSC m… Show more

“…Metabolomics, or metabolic profiling, is one of the younger "omics" technologies which comprehensively studies metabolic pathways in biological systems with the focus on metabolites [23,24]. Metabolomics encompasses high-throughput identification and elucidation of the small molecule metabolites that are produced by cells, tissues and microorganisms [25][26][27]. Metabolomics has previously been employed in attempts to diagnose epilepsy, however no single peripheral biomarker panel has demonstrated proven effectiveness [28][29][30][31].…”

“…Pulp-derived MSCs have been proposed for treating systemic disorders [87] and other types of MSCs, particularly in the area of neuroinflammatory and neurodegenerative diseases [103,104], whereas EVs have been advocated for the control and therapy of autoimmunity [105]. This promising outlook is certainly reinforced by progress in transcriptomics and single-cell analysis of MSCs [106,107], revealing different subsets and mechanisms of action. It is therefore not surprising that MSCs or their exosomes have recently been suggested as a treatment for severe COVID-19 [108][109][110][111].…”

“…Mesenchymal stem cell are being extensively investigated for their potential benefits in tissue engineering and regenerative medicine ( Sigmarsdottir et al, 2020 ). Of note, MSCs have many advantages, such as extensive sources, easy to culture in vitro , and less ethical issues, hence the study of MSCs is more extensive at present.…”
