Punicic acid is a conjugated linolenic acid with various biological activities including antiobesity, antioxidant, anticancer, and anti-inflammatory effects. It is often used as a nutraceutical, dietary additive, and animal feed. Currently, punicic acid is primarily extracted from pomegranate seed oil, but it is restricted due to the extended growth cycle, climatic limitations, and low recovery level. There have also been reports on the chemical synthesis of punicic acid, but it resulted in a mixture of structurally similar isomers, requiring additional purification/separation steps. In this study, a comprehensive strategy for the production of punicic acid in Yarrowia lipolytica was implemented by pushing the supply of linoleic acid precursors in a high-oleic oil strain, expressing multiple copies of the fatty acid conjugase gene from Punica granatum, engineering the acyl-editing pathway to improve the phosphatidylcholine pool, and promoting the assembly of punicic acid in the form of triglycerides. The optimal strain with high oil production capacity and a significantly increased punicic acid ratio accumulated 3072.72 mg/L punicic acid, accounting for 6.19% of total fatty acids in fed-batch fermentation, providing a viable, sustainable, and green approach for punicic acid production to substitute plant extraction and chemical synthesis production.

Microalgae have high lipid accumulation capacity, high growth rate and high photosynthetic efficiency which are considered as one of the most promising alternative sustainable feedstocks for producing lipid-based biofuels. However, commercialization feasibility of microalgal biofuel production is still conditioned to the high production cost. Enhancement of lipid accumulation in microalgae play a significant role in boosting the economics of biofuel production based on microalgal lipid. The major challenge of enhancing microalgal lipid accumulation lies in overcoming the trade-off between microalgal cell growth and lipid accumulation. Substantial approaches including genetic modifications of microalgal strains by metabolic engineering and process regulations of microalgae cultivation by integrating multiple optimization strategies widely applied in industrial microbiology have been investigated. In the present review, we critically discuss recent trends in the application of multiple molecular strategies to construct high performance microalgal strains by metabolic engineering and synergistic strategies of process optimization and stress operation to enhance microalgal lipid accumulation for biofuel production. Additionally, this review aims to emphasize the opportunities and challenges regarding scaled application of the strategic integration and its viability to make microalgal biofuel production a commercial reality in the near future.

A demanda energética cresce diariamente no mundo e é fortemente dependente dos combustíveis de origem fóssil, principais emissores de gases que provocam o efeito estufa. Impactos ambientais, como o aquecimento global, têm incentivado o desenvolvimento de combustíveis alternativos, no qual destaca-se os biocombustíveis, combustíveis obtidos do processamento de biomassa, uma fonte de energia renovável e sustentável. O biodiesel é um biocombustível produzido por transesterificação ou craqueamento de triglicerídeos oriundos de matérias-primas oleaginosas, o produto do craqueamento é denominado ecodiesel. O dendê é um oleaginoso que possui elevada produtividade de óleo, baixo custo e ausência de sazonalidade, porém o seu processamento gera uma grande quantidade de resíduos sólidos que necessitam de tratamento adequado. Desse modo, o objetivo deste trabalho foi apresentar uma revisão bibliográfica junto a uma proposta para a produção de ecodiesel a partir do dendê com a utilização da gaseificação para tratamento de resíduos sólidos, utilizando os conceitos de uma biorrefinaria. Um fluxograma de processo foi elaborado baseado em um levantamento teórico realizado em várias bases científicas, como SCIELO, Google Acadêmico e ScienceDirect e foi desenvolvido no software Lucidchart. Verificou-se que o dendê é promissor na produção de ecodiesel e que os processos de termoconversão, como a gaseificação, podem ser utilizados para agregar valor aos resíduos sólidos.
