Current and Potential Uses of Imiquimod

Abstract: Imiquimod, an imidazoquinoline amine, is an immune response modifier first FDA-approved for the treatment of external genital and perianal warts in 1997. Since its appearance on the market, its antiviral and antitumor properties have been used in the treatment of a variety of dermatologic conditions. In this review article, the basic mechanism of action of imiquimod, current FDA-approved and non-FDA-approved uses of imiquimod, and key points of medication application frequency, possible adverse effects, and us… Show more

“…Notably, on the basis of our results, R848 appears to exhibit its function not only in the priming phase of DCs by TSLP but also in the later phase of priming T cells by TSLP-DCs. In addition to these specialized roles of R848 for DCs in allergic responses, there may be a great advantage; that is, because imidazoquinoline has already been used to topically treat viral-mediated skin diseases by cream formulation (62)(63)(64)(65), this imidazoquinoline drug could be used without systemic side effects when R848 is clinically applied for the treatment of atopic dermatitis.…”

“…The mechanism of action of IQ is mainly related to the binding and stimulation of Toll-like receptors (TLRs), that are located on the surface of antigen-presenting cells, and serve to recognize and defend against invading pathogens (Chang et al 2005;Ceilley and Del Rosso 2006;. IQ is a potent TLR-7 agonist which induces synthesis and release of several endogenous pro-infl ammatory cytokines from Langerhans cells, monocytes/macrophages and dendritic cells.…”
