Curcumin Targeting NF-κB/Ubiquitin-Proteasome-System Axis Ameliorates Muscle Atrophy in Triple-Negative Breast Cancer Cachexia Mice

Abstract: Background. Curcumin is a polyphenol plant-derived compound with anti-inflammatory, antioxidant stress, and anticancer properties that make it have the potential to treat cancer cachexia. However, the role of it in breast cancer cachexia remains unclear. Methods. The 4T1 cells were subcutaneously injected into BALB/c mice to induce breast cancer cachexia. After tumor formation, the animals were divided into groups and given curcumin or saline interventions. The therapeutic effect of curcumin on breast cancer c… Show more

“…While a previous study investigating the effects of BC on muscle metabolism mainly focused on the soleus muscle ( 39 ), the gastrocnemius muscle was the primary focus in the present study because it represents the most common skeletal muscle associated with cachexia. Additionally, it is more involved in cancer cachexia pathogenesis, such as myogenesis and muscle atrophy, than the soleus muscle.…”

“…Inflammation is an essential cause of skeletal muscle atrophy [ 33 , 44 ]. NF-κB and the NF-κB-induced inflammatory factor TNFα can activate the transcription of murf1 and mafbx [ 40 , 45 , 46 ]. The mRNA levels of tnfα and il-6 were increased in the gastrocnemius muscle of adult rats fed alcohol [ 47 ].…”

“…In a randomized controlled trial, curcumin administration was associated with reduced circulating cytokine and creatine kinase levels in healthy adults with a lower perception of muscle soreness [ 253 ]. These effects depend on a direct effect of curcumin on skeletal muscle cells, improving post-exercise lactate accumulation [ 254 ], reducing protein breakdown [ 255 ], and stimulating protein synthesis through the sirtuin-3 pathway [ 256 ]. Furthermore, in older subjects, curcumin may promote the optimal microvascular perfusion of skeletal muscles [ 257 ].…”
