Curcumin Supplementation Lowers TNF-α, IL-6, IL-8, and MCP-1 Secretion in High Glucose-Treated Cultured Monocytes and Blood Levels of TNF-α, IL-6, MCP-1, Glucose, and Glycosylated Hemoglobin in Diabetic Rats

Abstract: This study examined the hypothesis that curcumin supplementation decreases blood levels of IL-6, MCP-1, TNF-alpha, hyperglycemia, and oxidative stress by using a cell-culture model and a diabetic rat model. U937 monocytes were cultured with control (7 mM) and high glucose (35 mM) in the absence or presence of curcumin (0.01-1 microM) at 37 degrees C for 24 h. Diabetes was induced in Sprague-Dawley rats by injection of streptozotocin (STZ) (i.p., 65 mg/kg BW). Control buffer, olive oil, or curcumin (100 mg/kg B… Show more

“…Our study showed that strong reduction in serum and tissue IL-6, with the use of combination treatment, more than each medication alone while curcumin alone showed a significant reduction than irbesartan and STZ group which was in agreement with the data obtained from Aggarwal and Harikumar [35] and Jain et al, [36]. The highly significant reduction observed with combination group may be explained by the presence of a synergistic effect between curcumin and irbesartan.…”

“…Curcumin, however, has been shown to down-regulate TNF expression (Jin et al, 2007;Lee et al, 2007;Zhang et al, 2008;. In addition to being induced by LPS, TNF is also upregulated by a variety of other stimuli including phorbol ester, palmitate and other inflammatory cytokines, and curcumin has been shown to block the expression of TNF induced by all of these stimuli (Abe et al, 1999;Lee et al, 2007;Jain et al, 2009;Wang et al, 2009).…”

“…[34][35][36] The main targets include the inflammatory transcription factor NF-jB, inflammatory enzymes (COX-2 and 5-lipoxygenase) and inflammatory cytokines such as TNFa, IL-1, IL-6 and IL-8. 36,37 Dietary curcumin has been shown to alleviate diabetes in high-fat diet-induced obese and leptin-deficient ob/ob mice, 36,37 as indicated by blood glucose levels, insulin tolerance and glycosylated hemoglobin. Mechanisms observed included reduced macrophage infiltration, increased adiponectin production in adipose tissue and decreased hepatic NF-jB activity.…”
