Curcumin Protects an SH-SY5Y Cell Model of Parkinson’s Disease Against Toxic Injury by Regulating HSP90

Abstract: Background/Aims: We aimed to explore the protective role of curcumin (Cur) in a cell model of Parkinson’s disease (PD) and its underlying mechanism. Methods: In this study, genes concerned with PD-related keywords were screened within DiGSeE database. The association network between Cur and selected genes was downloaded from STITCH, with the interactions analyzed by STRING. We built a mitochondrial toxin 1-methyl-4-phenylpyridinium (MPP+)-induced SH-SY5Y cell model of PD. Cell morphology was observe… Show more

“…We found that a-cyperone upregulated Bcl-2 and downregulated Bax in H 2 O 2 -treated SH-SY5Y cells, suggesting that a-cyperone suppressed mitochondrial dysfunction. Sanga et al have proved that the inhibition of cleaved-caspase-3 production protects a SH-SY5Y against toxic injury (Sang et al, 2018). In addition, Wu et al have reported that carnosic acid protects against 6-hydroxydopamine-induced neurotoxicity in in vivo and in vitro models of PD, by inducing antioxidative enzymes and inhibiting the production of cleavedcaspase-3 (Wu et al, 2015).…”

“…Besides, curcumin stimulates the migration of microglia to provide amyloid‐beta phagocytosis (Teter et al, 2019), leading to the alleviation of AD. To diminish the stress on neurons, curcumin induces heat shock protein 90 (HSP90), which is a key chaperone involved in the correct conformation of proteins, resulting in inhibition of apoptosis in dopaminergic neurons (Sang et al, 2018).…”

“…Naringin can lead to the anti-inflammatory pathway and provide neuroprotection in PD [70]. In in vitro PD model of SH-SY5Y cells, curcumin imparts a role in neuroprotection against toxic insult via controlling the HSP-90 [71]. Curcumin was studied in Pheochromocytoma Cell Line 12 (PC12) cells against 1-methyl-4-phenylpyridinium (MPP + ) toxicity and fixation of mitochondrial membrane potential was found against enhancement of intracellular ROS which leads to the instigation of iNOS, and over-expression of Bcl-2 [144].…”
