Curcumin as a permeability enhancer enhanced the antihyperlipidemic activity of dietary green tea extract

Abstract: Background Green tea has polyphenols like flavonoids and catechins; mainly epigallocatechin-3-gallate (EGCG), epicatechin gallate (ECG), epigallocatechin (EGC) and epicatechin (EC), out of which EGCG is of higher abundance. EGCG has shown preventive role in hypercholesterolemia. However, due to low oral bioavailability, a need arises to improve its membrane permeability and transporter-mediated intestinal efflux. Therefore, an attempt was made to enhance permeability and bioavailability of EGCG us… Show more

“…In the present study, the optimized formula was selected on the basis of maximum EE%, maximum Q 12h , and minimum PS. Additionally, the optimized EGCG-loaded spanlastic formula was validated by calculating % relative error via comparing the observed responses (Y1, Y2, and Y3) with their corresponding predicted values [23] as follows: % Relative error = (predicted value-observed value) × 100/predicted value (5) The optimized EGCG-loaded nanospanlastic formula was subjected to a comparative study with the corresponding niosomes and evaluated by additional characterization tests.…”

“…Catechins are the main polyphenolic constituents in green tea that are responsible for the majority of its anti-oxidant effects. Many studies have attributed the various valuable effects of green tea to (−) Epigallocatechin gallate (EGCG) [4,5]. EGCG is the most important green tea flavonoid and has attracted the interest of many researchers as a promising drug in the pharmaceutical, nutritional, and cosmetic fields.…”

“…Only a handful of studies have employed a dGTE formula [17,18,25,26], whereas positive benefits found elsewhere could be explained through the thermogenic effects of caffeine [11,19,29,30]. Whilst positive findings have been observed with a moderate EGCG dose (>300 mg•d −1 ), improving the bioavailability and effectiveness of GTE through standardised consumption away from food [20] and/or inclusion of antioxidant nutrients (e.g., quercetin, α-lipoic acid (LA), curcumin [31][32][33][34]) may lead to enhanced fat oxidation, body composition, and/or cardio-metabolic risk factor improvement, particularly in overweight individuals. Indeed, precedent for the approach of combining GTE with quercetin or LA has previously been observed in animal models [35,36].…”

“…Furthermore, recently developed elaborate computer algorithms (e.g., VIPER) could identify viable novel targets [https://www.sciencemag.org/news/2020/06/computer--algorithms--find--tumors--molecular--weak--spots]. In considering the possibility of adopting multiple complementary agents, attention should also be given to the avoidance of any antagonism [350].…”
