Curcumin Analog CH-5 Suppresses the Proliferation, Migration, and Invasion of the Human Gastric Cancer Cell Line HGC-27

Abstract: Gastric cancer is one of the most frequent malignant tumors in the world. The majority of patients are diagnosed with metastatic gastric cancer, which has a low survival rate. These data reinforce the importance of studying the anticancer activity of new molecules with the potential to suppress gastric cancer metastasis. Curcumin is a well-studied compound that has demonstrated anti-metastatic effects. Here we investigated if CH-5, a curcumin derivative compound, has anti-metastatic properties in the human gas… Show more

“…Transwell assays (Corning ® , Corning, NY, USA) were used to analyze the effect of trans -chalcone and licochalcone A on migration and invasion capacity of BT-20 cells, due to their high metastasis capacity. The assays were performed as described by Silva et al [ 32 ]. The treatments were realized using 30 µM and 15 µM for 24 h. Briefly, cells were maintained in serum-free medium overnight, and then 2 × 10 6 cells resuspended in 200 µL serum-free medium were seeded into the upper chamber of a 24-well Transwell plate with an 8 µm pore size (Corning ® ).…”

“…Next, 750 µL of culture medium containing different concentrations of D14 and D15 was added to the lower chamber together with a chemoattractant (10% FBS) and the plates were incubated for 24 h at 37 °C. Non-migrated cells on the upper side of the inserts were completely removed by swabbing, while the migrated cells attached to the bottom side of the Transwell membrane were fixed with paraformaldehyde and stained with Wright’s Giemsa solution [ 12 ]. For the invasion assay, the Transwell membranes were pre-coated with 0.75 mm of Matrigel (Corning ® ) according to the manufacturer’s instructions, and the same steps as in the migration assay were performed.…”

“…Our research group has been working to identify small molecules that can be used as metastasis inhibitors through the induction of p53 [ 12 , 13 ]. Prominent among these are chalcones, a group of polyphenolic compounds with an alpha-beta unsaturated ketone core moiety that exerts cytotoxic activity against different tumor cell lines through mechanisms such as cell cycle disruption, the inhibition of angiogenesis, and the induction of apoptosis [ 14 , 15 , 16 , 17 ].…”

“…Curcumin is shown to exhibit many anti-cancer effects through the inhibition of cell proliferation, promotion of cell apoptosis, prevention of tumor angiogenesis and metastasis, and the induction of autophagy [21][22][23][24][25].…”
