Culture of Shoot Apices of Theobroma cacao

Abstract: Surface sterilized buds of young cocoa plants (Theobroma cacao L.) taken at particular stages of the flush cycle were placed in Linsmaier and Skoog agar medium supplemented with a range of growth regulators. Only buds taken at the 1–2 (dormant) stage of the flush cycle and treated with gibberellic acid (GA3) alone and GA3plus kinetin (KN) supplement showed bud opening. In liquid Linsmaier and Skoog medium buds isolated at the 1–2 stage also responded to GA3 and KN. In this case addition of KN caused bud openin… Show more

“…Although efforts have been made to develop organogenesis-based propagation methods, cacao has proven to be recalcitrant to in vitro shoot regeneration and plant micropropagation (Orchard et al, 1979;Passey and Jones, 1983;Flynn et al, 1990;Figueira and Janick, 1994). Plant regeneration via somatic embryogenesis provides an alternative approach for clonal propagation of cacao.…”

“…There is a lack of consistency between inductive hormonal treatments with growth and development responses (Orchard et al, 1979;Passey and Jones, 1983;Janick and Whipkey, 1985;Blake and Maxwell, 1984;Legrand et at., 1984;Dufour and Dublin, 1985;Legrand and Mississo, 1986;Bertrand, 1987). “…This would enable new varieties to be introduced rapidly and virus-free stocks can be produced. Shoot tip culture (Orchard et al, 1979) has not yet been successful in cacao (Table 4). Callus and in some cases, callus with roots, has been successfully obtained from various tissues including cambium from mature stems (Archibald, 1954), seedling root, stem, hypocotyl, cotyledon (Hall and Collin, 1975), embryo and somatic tissue of anthers (Prior, 1977).…”
