Culture conditions influence satellite cell activation and survival of single myofibers

Abstract: Single myofiber isolation protocols allow to obtain an in vitro system in which the physical association between the myofiber and its stem cells, the satellite cells, is adequately preserved. This technique is an indispensable tool by which the muscle regeneration process can be recapitulated and studied in each specific phase, from satellite cell activation to proliferation, from differentiation to fusion. This study aims to clarify the effect of different culture conditions on single myofibers, their associa… Show more

“…These tissues can be utilized to measure contractile function (Brooks and Faulkner, 1988) and assess insulin-stimulated glucose uptake (Hansen et al, 1994). To overcome the short-term culture duration of intact muscle explants, single myofibers can be carefully dissected and isolated from the muscle belly and cultured for up to 7 days (Renzini et al, 2018), though most studies utilize 48-96 h time points. The single myofiber model is the gold-standard model to study satellite cell activation in vitro, due to SCs being retained within their niche and the ability to study SC dynamics with multiple modalities (Pasut et al, 2013;Wang Y. X. et al, 2019).…”

“…Single myofibers were isolated from EDL muscles of 4-6-week-old mice, as described [31][32][33]. Briefly, EDL muscles were dissected from tendon to tendon and incubated in DMEM (Sigma-Aldrich St. Louis, MS, USA) containing 0.2% collagenase I (Sigma-Aldrich) for 45 min.…”

“…Sodium benzoate is a common preservative added to processed meat products (Hoang and Vu, 2016). Furthermore, antibiotics are often added to the growth media of cells in long-term culture to prevent any infection from bacteria (Burton et al, 2000, Renzini et al, 2018, Stephens et al, 2018. Patents for industrial production of cell-based meat state that the process will be achieved without antibiotics (or hormones) (Van Eelen, 2007).…”
