Cultural and Physiological Observations On Trypanosoma Rhodesiense and Trypanosoma Gambiense

Abstract: 1. A diphasic medium of simple preparation is described for the indefinite cultivation of T. rhodesiense and T. gambiense. 2. The chief advantage of the medium is that it contains rabbit blood and thus obviates the necessity of using human blood. 3. The flagellates develop only to the proventricular stage; hence the cultures are noninfective. 4. The proventricular forms of both T. rhodesiense and T. gambiense consume sugar with the concomitant formation of acid. They are aerobic fermenters. 5. Very little, if … Show more

“…Informed consent was obtained from patients or guardians, and ethical clearance was granted by the institutional review boards in Belgium and Nepal. Briefly, bone marrow aspirates were directly inoculated into Tobie's blood agar medium at ¡26 xC for the cultivation of parasites (Tobie et al 1950). 1) : 6 endemic districts with a high rate of VL transmission during the past, and the non-endemic hilly district Bhojpur from which a number of cases were reported.…”

“…Parasite promastigotes were derived from bone marrow aspirates of VL patients by culturing in Tobie’s blood agar medium with Locke’s overlay [13], with 200 IU/ml penicillin and 200 μg/ml streptomycin. Once the parasites were fully grown from the clinical material, they were transferred to M199 (Sigma-Aldrich, cat.…”

“…The earliest method of in vitro testing of drugs on African trypanosomes relied heavily on the use of Tobie's medium, which is a biphasic culture method that involves transfer of the parasite cells into a secondary medium requiring the addition of heat‐inactivated defibrinated blood. Effect of the drugs was evaluated microscopically . This has been replaced by the use of simpler and more economic monophasic culture method in which the parasites are maintained in all‐liquid medium, Hirumi's modified Iscove's medium 9 supplemented with 10% fetal bovine serum, at 37°C and in a 5% CO 2 atmosphere .…”
