Abnormal vertical growth (AVG) in macadamia is a syndrome that reduces flowering and alters tree architecture, resulting in significant yield losses. The epidemiological parameters suggest the cause is probably a biotic agent with the ability to modulate plant hormone production or signalling. To determine if a fungal pathogen is the cause of AVG, we compared the fungal profile of macadamia trees with or without AVG symptoms using conventional culturing and DNA metabarcoding techniques. A diverse range of fungi was isolated from leaf, root and stem samples of macadamia cultivars HAES 344 and A16. The abundance of the isolated fungi was at least three‐fold higher in trees with AVG symptoms than in trees at the non‐AVG site. Similar results were obtained using DNA metabarcoding. Although the fungal genus Sebacina was the most dominant in the DNA metabarcoding, whereas Diaporthe (>40%) was dominant using the culturing techniques, no unique fungal species was consistently associated with all AVG samples. Therefore, we conclude that it is unlikely that the cause of AVG in macadamia is a fungal pathogen.

Macadamia (Macadamia integrifolia) trees are an important source of revenue in rainforest ecosystems. Their nuts are rich in vitamins, minerals, fiber, antioxidants, and monounsaturated oils. The fungus Lasiodiplodia theobromae, however, is a major disease problem, causing kernel rot and other disease symptoms. In the present study, a dual confrontation assay was used to evaluate the inhibitory effect of an endophytic strain of Trichoderma hamatum C9 from macadamia root against L. theobromae. Volatiles and cell-free culture filtrate of T. hamatum were also used to assess their antifungal activity against L. theobromae. Results suggested that T. hamatum exhibited a significant inhibitory effect against L. theobromae in vitro. Further results of a biocontrol assay indicated that a spray treatment of T. hamatum conidial suspension significantly decreased the size of lesions caused by artificially inoculated L. theobromae on macadamia leaves, as well as the disease index in young trees inoculated with L. theobromae, relative to sterile water controls. Collectively, our findings indicate that T. hamatum C9 represents a potential biocontrol agent that can be used to manage L. theobromae on macadamia.

The availability of limiting nutrients plays a crucial role in shaping communities of endophytes. Moreover, whether fungal endophytes are host-specific remains controversial. We hypothesized that in a harsh and nitrogen (N)-deficient area, diversity and community composition of foliar endophytic fungi (FEFs) varied substantially among plots with experimentally elevated levels of macronutrients, and thus, N availability, instead of host species identity, would have a greater influence in structuring fungal communities at different scales. We also expected an important subset of taxa shared among numerous host species and N gradients to form a community-wide core microbiome. We measured the leaf functional traits and community structures of FEFs of three commonly seen species in an alpine meadow nested with a long-term N fertilization experiment. We found that host plant identity was a powerful factor driving the endophytic fungal community in leaves, even in habitats where productivity was strongly limited by nitrogen (p < 0.001). We also found that within the same host, nitrogen was an important driving force for the composition of the endophytic fungi community (p < 0.05). In addition, the leaf carbon content was the most important functional trait that limited the diversity of endophytic fungi (p < 0.001). Finally, we documented a distinct core microbiome shared among our three focal species and N gradients.
