A novel, efficient, and accurate fingerprinting method using high performance liquid chromatography-photodiode array detection has been developed and optimized for the investigation and demonstration of the variance in chemical properties among Siraitia grosvenorii fruits from different origins. The effects of growth stages, cultivated varieties, collection locations, and fruit portions of the herb on chromatographic fingerprints were examined. Eleven compounds were identified on chromatograms by comparing the retention time and UV spectrum of each peak separately with those of external references. The results revealed that chromatographic fingerprints, combining similarity or hierarchical clustering analysis along with reference compounds, could efficiently identify and distinguish S. grosvenorii fruits from different sources, which provided helpful clues for studying the plants' secondary metabolites and benefitted quality control.

Siraitia grosvenorii (Swingle) C. Jeffrey, a member of the family Cucurbitaceae, is a unique economic and medicinal plant grown in China. For more than 300 years, S. grosvenorii has been used as a natural sweetener and as a traditional medicine for the treatment of pharyngitis, pharyngeal pain, as well as an anti-tussive remedy in China. It is one of the first approved medicine food homology species in China. It has been widely studied as a natural product with high development potential. Therefore, the present paper provides a review of the botanical characterization, traditional uses and ethnopharmacology, food and nutritional values, chemical constituents, pharmacological effects, toxicology, and development direction for the future of S. grosvenorii. Phytochemical studies have revealed that the chemical composition of this plant mainly includes iridoid and phenylpropanoid glycosides. Several compounds such as triterpenoids, flavonoids, and amino acids have been isolated from the plant. S. grosvenorii and its active constituents possess broad pharmacological properties, such as antioxidant, hypoglycemic, immunologic, anti-tussive and sputum-reducing, hepatoprotective, and antimicrobial activities, etc. By documenting the comprehensive information of S. grosvenorii, we hope to establishes the groundwork for further research on the mechanism of action of S. grosvenorii and its development as a new health food in the future.

Siraitia fructus, known as Luo-Han-Guo (LHG) in Chinese, shows good antidiabetic activity. Gut microbiota have emerged as parameter of the pathophysiology of type 2 diabetes (t2D) mellitus. In this study, in vitro biotransformation of LHG extract by t2D and normal human intestinal microbiota was analyzed using ultraperformance liquid chromatography with time-of-flight mass spectrometry. A total of 19 and 12 metabolites were characterized in t2D and normal human intestinal microbiota, respectively. These metabolites were formed by a series of metabolic reactions including deglycosylation, oxidation, isomerization, and/or deoxidation. The metabolite classes of LHG extract in normal and t2D human intestinal microbiota were nearly the same, but oxidation and isomerization reactions were usually present in t2D human intestinal bacteria. This might be due to metabolic activation by pathological human intestinal bacteria. Our study indicates that there are differences in the biotransformation of LHG extract in normal and t2D human intestinal bacteria.
